Direct Preparation of Indole Hemiaminals through Organocatalytic Nucleophilic Addition of Indole to Aldehydes

Hemiaminals are common in natural products as well as bio-active compounds. Hemiaminals with an indole moiety are particularly attractive due to the significant bioactivity of indoles. Herein, we reported an efficient organocatalyzed indole N-1 nucleophilic addition of alpha-oxoaldehydes to deliver various indole hemiaminals in good yields (up to 92%) and excellent regioselectivities with DABCO or triethylamine as the catalyst. The method is characterized by mild reaction conditions, widely available reagents, and general substrate scope, and it is also applicable to late-stage transformations without affecting the hemiaminal group. In addition, we carried out this reaction in an enantioselective fashion in good yields and high ee values with two general substrates.

Encapsulation of nodal segments of Allamanda cathartica for short-term storage and germplasm exchange

Synthetic seed has the potential to be used as a cost-effective tool for propagation and storage of germplasm. In the present study, synthetic seeds were produced by encapsulating nodal segments of Allamanda cathartica in calcium alginate gel. The best gel complex was achieved using 3% sodium alginate and 100 mM calcium chloride (CaCl2 center dot 2H(2)O). Maximum conversion response of encapsulated nodal segments into plantlets was obtained on Murashige and Skoog (MS) medium with 3.0 mu M meta-Topolin (mT) and 0.5 mu M alpha-naphthalene acetic acid (NAA). Encapsulated nodal segments could be stored at low temperature (4 degrees C) up to 6 weeks with a survival frequency of 85%. The regenerated shoots rooted on half-strength MS medium containing 0.5 mu M indole-3-butyric acid (IBA). Plantlets with adequate root and shoot systems were hardened off successfully with 90% success rate. Inter Simple Sequence Repeat (ISSR) markers were used to evaluate genetic stability among synthetic seed derived plantlets. The repeated high regeneration of plants from alginate-coated axillary buds and apparently good viability of the synseeds after four weeks of storage indicate that this is a viable strategy for the exchange of A. cathartica genetic resources. Mycelial culture extracts of selected wood-decay mushrooms as a source of skin-protecting factors

Objectives This study analyzed the content of substances with cosmetologic properties in the extracts obtained from the mycelial cultures of Ganoderma applanatum, Laetiporus sulphureus, and Trametes versicolor. The effect of these extracts on the inhibition of tyrosinase and hyaluronidase was determined, and their values of sun protection factor (SPF) were calculated. Results The total amount of phenolic acids in the extracts ranged from 2.69 (G. applanatum) to 10.30 mg/100 g dry weight (T. versicolor). The total amount of sterols was estimated at 48.40 (T. versicolor) to 201.04 mg/100 g dry weight (L. sulphureus), and that of indoles at 2.90 (G. applanatum) to 16.74 mg/100 dry weight (L. sulphureus). Kojic acid was determined in the extracts of L. sulphureus and G. applanatum. It was observed that L. sulphureus extract caused dose-dependent inhibition of hyaluronidase, while all the extracts inhibited tyrosinase. The extract of G. applanatum exhibited an SPF value of similar to 9. Conclusions The results showed that the mycelial cultures of the studied species may be used as an alternative source of substances used in cosmetology.

Application of gas sensors for modelling the dynamic growth of Pseudomonas in pork stored at different temperatures

Pseudomonas have a faster growth rate over other bacteria in chilled meat under aerobic conditions. A non-destructive method for modelling the dynamic growth of Pseudomonas in pork stored at different temperatures using gas sensors was presented in our work. Based on selected gas sensor data, the first-order kinetic equations (Gompertz and Logistic Functions) combined with the secondary model (Square-root Function) effectively simulated Pseudomonas growth in pork at different temperatures with R-2 and RMSE values of 0.71-0.97 and 0.27-0.84, respectively. Additionally, these models showed high accuracy with correlation coefficients greater than 0.90, in addition to several individual accuracy values. Furthermore, HS-SPME/GC-MS results demonstrated the presence of identified key volatiles in samples inoculated with Pseudomonas, including three amine compounds (mercaptamine, 1-octanamine and 1-heptadecanamine), phenol and indole. Our work showed that gas sensors are a rapid, easy and non-destructive method with acceptable feasibility in modelling the dynamic growth of spoilage microorganisms in meat.

Biodiversity of Convolvulaceous species that contain ergot alkaloids, indole diterpene alkaloids, and swainsonine

Convolvulaceous species have been reported to contain several bioactive principles thought to be toxic to livestock including the calystegines, swainsonine, ergot alkaloids, and indole diterpene alkaloids. Swainsonine, ergot alkaloids, and indole diterpene alkaloids are produced by seed transmitted fungal symbionts associated with their respective plant host, while the calystegines are produced by the plant. To date, Ipomoea asarifolia and Ipomoea muelleri represent the only Ipomoea species and members of the Convolvulaceae known to contain indole diterpene alkaloids, however several other Convolvulaceous species are reported to contain ergot alkaloids. To further explore the biodiversity of species that may contain indole diterpenes, we analyzed several Convolvulaceous species (n = 30) for indole diterpene alkaloids, representing four genera, Argyreia, Ipomoea, Stictocardia, and Turbina, that had been previously reported to contain ergot alkaloids. These species were also verified to contain ergot alkaloids and subsequently analyzed for swainsonine. Ergot alkaloids were detected in 18 species representing all four genera screened, indole diterpenes were detected in two Argyreia species and eight Ipomoea species of the 18 that contained ergot alkaloids, and swainsonine was detected in two Ipomoea species. The data suggest a strong association exists between the relationship of the Periglandula species associated with each host and the occurrence of the ergot alkaloids and/or the indole diterpenes reported here. Likewise there appears to be an association between the occurrence of the respective bioactive principle and the genetic relatedness of the respective host plant species.
