The derived 3D-QSAR models showed acceptable r(2) (0.91) and q(2) (0.91) with a root mean squared error (RMSE) of 0.08. The models were cross-validated using the leave-one-out method. Applying the same QSAR model to another congeneric series of amphiphilic indoles externally validated the QSAR model. The model could appreciably predict the activity (pMIC(50)) of this congeneric series of amphiphilic indoles, with an RMSE of 0.49, indicating the robustness of the model and its efficiency in predicting the potentially active compounds.

Indole and coumarin are the most versatile and cogent heterocyclic scaffolds which are not only decisively used in the synthesis of various organic compounds but also play a consequence role in natural product synthesis, modulation of biofilm formation, virulence and stress responses. In this review, we provide an overview of the synthesis of coumarin and indoles and their pharmacological evolution. The coumarin was synthesized using phenols, salicylaldehyde, benzaldehyde, phenylacetate, styrenes, and cinnamic acid with different catalyst and photocatalyst to give the best yield. Similarly, Indoles were synthesized iodobenzoic acid, alkynes, amines, alcohols, Nitrobenzaldehyde with the different catalyst under conventional and irradiation method. The effect of various catalytic medium, solvents and operational condition are discussed for obtaining the best yield. A comparative account of various reaction pathways like one-pot synthesis (Multicomponent reaction) and the multistep reaction of coumarin and indoles are discussed.

Indoles, including 5-methoxy-2-methyl-indole (5MeO2MeIn), are a potent trigger of strobilation in jellyfish polyps. Indoles may be an alternative method to cooling to induce strobilation, but the ephyrae produced should have similar or better rates of survival, deformities, growth, and behaviour to those produced by cooling, and polyps should remain viable after strobilating. We used two experiments to optimise the use of 5MeO2MeIn to induce strobilation in Aurelia coerulea (Scyphozoa). First, we compared rates of strobilation, survival, and deformities of ephyrae and budding of polyps exposed to 1.25, 2.5, and 5.0 mu M 5MeO2MeIn for 3 days to cooling at 14 degrees C. Polyps in the indole and cooled treatments strobilated after 10 days and produced similar numbers of ephyrae, but more ephyrae were deformed in the 5.0 mu M treatment and survival of ephyrae was lower in the 2.5 and 5.0 mu M treatments than the cooled treatment. Polyps exposed to all three concentrations of indoles failed to bud or died after strobilation. Next, we exposed polyps to 0.7, 1.25, and 2.5 mu M 5MeO2MeIn for four hours. Polyps exposed to all indole concentrations strobilated and produced similar numbers of ephyrae, but more ephyrae were deformed in the 2.5 mu M treatment. Survival, behaviour, and sizes of ephyrae were similar 7 and 14 days after strobilation (although ephyrae pulsed faster in the 1.25 mu M treatment at 14 days) and budding rates were similar in the indole and cooled treatments. Thus, exposing polyps to 0.7-1.25 mu M 5MeO2MeIn for 4 h is a viable and efficient alternative to cooling to induce strobilation in polyps.

ERECTAs are receptor-like kinases that regulate plant biomass and stress resistance. In this study, the wheat (Triticum aestivum) TaERECTA gene was used as a probe to identify the SbERECTA family genes (SbERs) in the sorghum (Sorghum bicolor) genome, analyse their subcellular localisation and characterise their expression. Results showed that the two SbER members, SbER10 with three copies (SbER10_X1, SbER10_X2, and SbER10_X3) and SbER4 with two copies (SbER4_X1 and SbER4_X2), were found on chromosomes 10 and 4 of sorghum, respectively. SbER10 had the highest expression level in the pedicel tissue and showed a remarkable response under treatment with abscisic acid, brassinolide, gibberellin and indole-3-acetic acid. SbER10_X1, functioning on the cell membrane and chloroplast, exhibited abundant transcript in only a few sorghum varieties that are grown in mountainous areas and receive strong light, heat, and water supply. Expression of SbER10_X1 was significantly and positively correlated with plant biomass of 32 sorghum germplasm resources. These results indicate that SbER10 genes have an important regulatory role in sorghum growth, and increasing SbER10 transcription level offers a potential strategic target for breeding or biotechnological approaches to enhance sorghum biomass and environmental adaptability.
