A cobalt-complex-catalyzed borylation of a wide range of alkyl halides with a diboron reagent (B(2)pin(2) or B(2 )neop(2)) has been developed under mild reaction conditions, demonstrating the first cobalt-mediated cross-coupling with alkyl electrophiles. This protocol allows alkyl boronic esters to be accessed from alkyl halides, including alkyl chlorides, which were used rarely as coupling partners. Mechanistic studies reveal the possible involvement of an alkyl radical intermediate in this cobalt-mediated catalytic cycle.

Silver and palladium nanoparticles anchored to the core-shell Fe3O4@SiO2@Al2O3 were successfully synthesized based on MIL(Al)96 MOF. The nanocomposites, 15-20 nm in size, exhibited high catalytic activity for selective aerobic oxidation of alcohols (even aliphatic alcohols) to the corresponding carbonyl compounds at room temperature without addition of any other additives. In this regard, the Fe3O4@SiO2@Al2O3/Pd,Ag exhibited more catalytic activity than the others. The cytotoxic effect of nanocomposites was tested in-vitro against MCF-7 human breast cancer cells by MTT assay. The results showed that all synthesized nanocomposites will be promising and potential as anticancer agents, with sample Fe3O4@SiO2@Al2O3 being the most cytotoxic after 24 and 48 h. Also, the obtained nanocomposites could induce apoptosis in breast cancer cell line (MCF-7) and the ability of induce apoptosis was in the following order: Fe3O4@SiO2@Al2O3@Ag > Fe3O4@SiO2@Al2O3 > Fe3O4@SiO2@Al2O3@Pd,Ag > Fe3O4@SiO2@Al2O3@Pd.

Binary organocatalytic ring-opening polymerization (ROP) of N-Boc-1,4-oxazepan-7-one (OxP(BOC)) is proposed as a versatile method to generate metal-free poly(amine-co-ester)s and the corresponding biodegradable polycationic homo/block (co)polymers. Optimization experiments with different organocatalysts, including 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU), 1,5,7-triazabicyclo[4.4.0]dec-5-ene (TBD), DBU/1-(3,5-bis(trifluoromethyl)phenyl)-3-cyclohexylthiourea (TU1), and phosphazene/thiourea, revealed that t-Bu-P-4/TU1 leads to a fast and controlled/living ROP with benzyl alcohol (BnOH) as an initiator and thus allows the formation of well-defined poly(amine-co-ester)s with high molecular mass (M-n,M-SEC up to 38.8 kg mol(-1)) and narrow dispersity. Well-defined poly(N-Boc-1,4-oxazepan-7-one)-based diblock copolymers were also synthesized successfully via the controlled/living ROP, regardless of whether hydrophilic poly(ethylene glycol) (PEG) or hydrophobic polycaprolactone (PCL) was used as a macroinitiator. After treatment with trifluoroacetic acid, the corresponding water-soluble polycationic homopolymer, as well as the hydrophilic (PEG -based copolymer) and amphiphilic (PCL-based copolymer) polycationic diblock copolymers, was obtained. Dynamic light scattering in water and transmission electron microscopy analyses revealed that both polycationic copolymers self-assembled spontaneously to nanostructures; the hydrophilic block copolymer formed spherical nanoparticles, while the amphiphilic one formed rod-shaped nanoparticles.

Multidrug resistance (MDR) is a major obstacle for effective chemotherapeutic treatment of cancer frequently leading to failure of the therapy. MDR is often associated with the overexpression of ABC transport proteins like ABCB1 or ABCG2 which efflux harmful substances out of cells at the cost of ATP hydrolysis. One way to overcome MDR is to apply potent inhibitors of ABC transporters to restore the sensitivity of the cells toward cytostatic agents. This study focusses on the synthesis and evaluation of novel 2,4-disubstituted quinazoline derivatives regarding the structure-activity-relationship (SAR), their ability to reverse MDR and their mode of interaction with ABCG2. Hence, the inhibitory potency and selectivity toward ABCG2 was determined. Moreover, the intrinsic cytotoxicity and the reversal of MDR were investigated. Interaction type studies with the substrate Hoechst 33342 and conformational analyses of ABCG2 with 5D3 monoclonal antibody were performed for a better understanding of the underlying mechanisms. In our study we could further enhance the inhibitory effect against ABCG2 (compound 31, IC50: 55 nM) and identify the structural features that are crucial for inhibitory potency, the impact on transport activity and binding to the protein. An efficient method for the synthesis of a new highly substituted pyrrolidinone derivative is developed via a three-component one-pot reaction of aniline, diethyl acetylenedicarboxylate, and 4-anisaldehyde. The structure of ethyl 4-hydroxy-2-(4-methoxyphenyl)-5-oxo-1-phenyl-2,5-dihydro-1H-pyrrole-3-carboxylate is confirmed by FTIR, H-1 and C-13 NMR spectroscopy. Unambiguous evidence for the structure of the synthesized product is obtained from the single crystal X-ray analysis. The comparison of both its molecular structure and intermolecular interactions with the previously known analogous compounds reveals that the mutual syn- or antiperiplanar orientation of ester C=O and pyrrolinone-bound OH is related to the type of molecular dimers formed in the solid state. The geometrical structure, the HOMO-LUMO analysis (electrophilicity index), H-1 and C-13 chemical shifts of the desired product are also calculated by DFT at the B3LYP level with the 6-31++G(d,p) basis set. The calculated chemical shifts are in compliance with the experimental data.

We report a new strategy for the conversion of carboxylic acids into potent acid triflate electrophiles. The reaction involves oxidative carbonylation of carboxylic acids with I-2 in the presence of AgOTf, and is postulated to proceed via acyl hypoiodites that react with CO to form acid triflates. Coupling this chemistry with subsequent trapping with arenes offers a mild, room temperature approach to generate ketones directly from broadly available carboxylic acids without the use of corrosive and reactive Lewis or Bronsted acid additives, and instead from compounds that are readily available, stable, and functional group compatible.

The floral fragrance of plants is an important indicator in their evaluation. The aroma of sweet cherry flowers is mainly derived from their essential oil. In this study, based on the results of a single-factor experiment, a Box-Behnken design was adopted for ultrasound- and microwave-assisted extraction of essential oil from sweet cherry flowers of the Brooks cultivar. With the objective of extracting the maximum essential oil yield (w/w), the optimal extraction process conditions were a liquid-solid ratio of 52 mL g(-1), an extraction time of 27 min, and a microwave power of 435 W. The essential oil yield was 1.23%, which was close to the theoretical prediction. The volatile organic compounds (VOCs) of the sweet cherry flowers of four cultivars (Brooks, Black Pearl, Tieton and Summit) were identified via headspace solid phase microextraction (SPME) and gas chromatography-mass spectrometry (GC-MS). The results showed that a total of 155 VOCs were identified and classified in the essential oil from sweet cherry flowers of four cultivars, 65 of which were shared among the cultivars. The highest contents of VOCs were aldehydes, alcohols, ketones and esters. Ethanol, linalool, lilac alcohol, acetaldehyde, (E)-2-hexenal, benzaldehyde and dimethyl sulfide were the major volatiles, which were mainly responsible for the characteristic aroma of sweet cherry flowers. It was concluded that the VOCs of sweet cherry flowers were qualitatively similar; however, relative content differences were observed in the four cultivars. This study provides a theoretical basis for the metabolism and regulation of the VOCs of sweet cherry flowers.

Silyl-substituted ketenes are attractive molecular building blocks due to their stability and ease of storage, as opposed to unstable alkyl and aryl ketenes. To better understand the reactivity of silyl ketenes and, in turn, their use in the preparation of highly functionalized small molecules, the reaction of silyl ketenes with different nucleophiles was studied. The addition of alcohol, amine, or thiol nucleophiles to the central carbon of silyl ketene, followed by proton transfer afforded alpha-silyl ester, amide, or thio-ester, respectively. Catalytic amounts of Lewis acid greatly increase the rate of the reaction, and the impact of nucleophile, Lewis acid, and silyl substituent are evaluated. The small molecules produced from these reactions give insight into the use of silyl ketenes as building blocks for complex molecular structures.

Polymer functionality greatly determines many of the key properties of these materials, such as glass-transition temperature, electrical and thermal conductivity, thermal stability, mechanical strength, and processability. Despite the importance of polymer functionality in determining material properties, the synthesis of functional polymers, with well-defined molecular weights and compositions, can still present a significant challenge, with many of the methods related to pre- or postpolymerization modification lacking synthetic scope, or requiring harsh functionalization conditions or transition-metal coupling reactions to install the desired functionality. Perfluoroaromatic systems are promising for the preparation of novel polymer architectures given that they can be readily functionalized using simple nucleophilic chemistries under very mild basic conditions. While promising, these systems have displayed some drawbacks. Previous work has shown that perfluoroaromatics, such as perfluoropyridine, can demonstrate a high degree of chemical reversibility with heteroatom nucleophiles. If the synthetic potential of these systems is to be realized, then a strategy for the rational design of stable monomers must be developed. Herein, we report the design, synthesis, and characterization of a series of unexplored heteroatom-based ring-opening metathesis polymerization (ROMP)-active monomers containing a reactive perfluoropyridine pendent group, which can be used to readily prepare a wide variety of aryl ether-functionalized polymers, using both pre- and postpolymerization modification strategies. We also establish a direct connection between the dihedral angle of the monomer and its propensity to undergo reversible addition reactions, establishing functional criteria for the design of pre- and postmodifiable systems.

An efficient and green method, combining enzymatic and visible-light catalysis for synthesis of the widely applicable 2-substituted benzothiazoles, has been developed. This method features a relay catalysis protocol consisting of biocatalytic promiscuity and visible-light-induced subsequent oxidization of 2-phenyl benzothiazolines. The whole reaction process is very high-efficiency, achieving 99% yield in just 10 min, under an air atmosphere, nearly 100% atomic utilization, and the 2-substituted benzothiazole products were obtained in good to excellent yields with a wide range of substrates. This reaction is the other example of combining the nonnatural catalytic activity of hydrolases with visible-light catalysis for organic synthesis and the catalytic system does not require additional oxidants or metals, which is good for the environment.
