Proprotein convertase subtilisin/kexin type 9 (PCSK9) is a secreted protein and its deficiency markedly enhanced the survival rate of patient with cardiovascular diseases (CVDs). Forty berberine (BBR) derivatives were synthesized and evaluated for their activities on down-regulating the transcription of PCSK9 in HepG2 cells, taking BBR as the lead. Structure-activity relationship (SAR) analysis revealed that 2,3-dimethoxy moiety might be beneficial for activity. Among them, 9k displayed the most potent activity with IC50 value of 9.5 +/- 0.5 mu M, better than that of BBR. Also, it significantly decreased PCSK9 protein level at cellular level, as well as in the liver and serum of mice in vivo. Furthermore, 9k markedly increased LDLR expression and LDL-C clearance via downregulating PCSK9 protein. The mechanism of action of 9k is targeting HNF1 alpha and/or Sp1 cluster modulation upstream of PCSK9, a different one from BBR. Therefore, 9k might have the potential to be a novel PCSK9 transcriptional inhibitor for the treatment of atherosclerosis, worthy for further investigation.

3,4,7,8-tetrahydro-3,3-dimethyl-11-aryl-2H-pyridazino[1,2-a]indazole-1,6,9(11H)-triones 5a-j were synthesized by a one-pot cyclocondensation of four components such as succinic anhydride, hydrazine hydrate, benzaldehyde derivatives and dimedone at room temperature through a multi-component reaction and solvent-free condition by using 1-methyl imidazolium trinitromethanide {[HMIM]C(NO2)(3)} as a very effective ionic liquid catalyst. All synthesized compounds which are soluble in DMSO were subjected to antibacterial activity assay against Escherichia coli, Salmonella enteritidis, Staphylococcus aureus, and Bacillus cereus. Results showed that the compounds had antibacterial activity against some bacteria tested. In addition, the antioxidant activity of the compounds was evaluated by DPPH free radical scavenging assay. Our results indicated that most of the compounds had excellent dose-dependent antiradical properties. Based on obtained data, the compounds possessed moderate to excellent bactericidal effect on both Gram-positive and Gram-negative bacteria. Moreover, the compounds 5b and 5g exhibited the most anti-proliferative potencies against MCF-7 (breast carcinoma), A549 (non-small lung carcinoma) and AGS (gastric carcinoma) cell lines.

Characterization of volatile off-odor compounds in thermal duck egg gels was investigated by gas chromatography-olfactometry-mass spectrometry (GC-O-MS), odor activity values (OAVs) and aroma recombination. The results demonstrated that the off-odor of thermal duck egg mainly came from egg yolk (EY), and content of aldehydes was the highest in EY. Anaerobic treatment in nitrogen contributed to decreased off-odor sensory scores and volatile components of the heated egg gels. Aerobic EY gels (treated in air) were determined to contain eight volatile compounds with OAVs greater than 1, such as hexanal, nonanal, benzaldehyde, (E)-2-octenal, (E)-2-nonenal, (E, E)-2,4-decadienal, 1-octen-3-ol and 2-pentylfuran. However, three key volatile components (nonanal, benzaldehyde and (E)-2-nonenal) with OAVs greater than 1 were found in anaerobic EY gels. Moreover, on the basis of results of recombination and omission experiments, hexanal, (E)-2-octenal, (E, E)2,4-decadienal, 1-octen-3-ol and 2-pentylfuran were identified as key off-odor components in thermal EY gels. Sensory evaluation of the recombination model with five aroma compounds scored 4.38 out of 5 points. The major aroma profile of the off-flavor from egg EY gels included strong fatty, nutty, and mushroom aromas.

A catalytic intermolecular amination of nonactivated tertiary C(sp(3))-H bonds (BDE of 96 kcal. mol(-1)) is reported for substrates displaying an activated benzylic site (BDE of 85 kcal.mol(-1)). The tertiary C(sp(3))-H bond is selectively functionalized to afford alpha, alpha, alpha-trisubstituted amides in high yields. This unusual site-selectivity results from the synergistic combination of Rh-2(Stfpttl)(4), a rhodium(II) complex with a well-defined catalytic pocket, with tert-butylphenol sulfamate (TBPhsNH(2)), which leads to a discriminating rhodium-bound nitrene species under mild oxidative conditions. This catalytic system is very robust, and the reaction was performed on a 50 mmol scale with only 0.01 mol % of catalyst. The TBPhs group can be removed under mild conditions to afford the corresponding NH-free amines.

A series of hybrids 10a-v based on coumarin/pyrazole oxime have been synthesized, and exhibit good to excellent antitumor activities. Compound 10n has shown remarkable anticancer effect on SMMC-7721 cells (IC50 = 2.08 mu M), which is considerably lower than 5-FU (IC50 = 37.8 mu M) and similar to ADM (IC50 = 2.67 mu M), with little effect on normal hepatic cells LO2. Notably, the suppression experiments of metastatic activities reveal that 10n also displays significant anti-metastasis effects through inhibiting cell migration and invasion in highly metastatic SMMC-7721 cell line, and dose-dependently reverses TGF-beta 1-induced epithelial-mesenchymal transition (EMT) procedure better than ADM. Finally, 10n also possesses low acute toxicity and potent tumor growth inhibitory property against SMMC-7721 cell lines in vivo. Cuprous halides, best described as (CuX)(n) (X = Cl-, Br-, and I-) in their solid state, catalyse selective aerobic oxidation of alcohols with the assistance of both NMI (N-methylimidazole) and TEMPO (2,2,6,6-tetramethylpiperidine-1-oxyl), and the iodide generally demonstrates the highest activity, for example, in the oxidation of 1-octanol at ambient temperature under 24 h’ reaction. However, in the aerobic oxidation of benzylic alcohols, the chloride showed superiority to the iodide in that the aerobic oxidation was quantitatively completed within 3 h at ambient temperature whereas the iodide showed only about half the activity of the chloride analogue. By probing the system using electrochemistry, electric conductivity, and H-1 NMR titration, it was revealed that the surprising anomaly was due to the difference in the rate of forming active species, [Cu (NMI)(2)X(MeCN)], from the polymeric solid in a two-stage process. Substrates expansion of 11 benzylic alcohols indicated that CuCl/NMI/TEMPO system demonstrated quantitative conversion of benzylic alcohols into corresponding aldehydes within 3 h and showed great tolerance to the substituents on the phenyl ring of the substrates. Furthermore, electron-withdrawing substituent was beneficial to the oxidation and could offset the steric effect at orthro-substituent. Such a behaviour suggested that in the catalysis, increasing the acidity of the hydroxyl group (OH) of the substrates could ease the oxidation, which implied that the deprotonation via an internal pathway might be one of the rate-determining steps. Our results also showed that the anion halide participated actively in the catalysis by coordinating to Cu(I) in the active species.

The intramolecular [4+2] cycloaddition between arenes and allenes first reported by Himbert gives rapid access to rigid polycyclic scaffolds. Herein, we report a one-pot oxyalkynylation/cycloaddition reaction proceeding under mild conditions (23-90 degrees C) and providing complex polycyclic architectures with high efficiency, and atom and step economy. The bicyclo[2.2.2]octadiene products were obtained with a wide variety of useful functional groups and were successfully applied as chiral ligands for metal catalysis. Polydopamine/MIL-53(Fe) (PDA/MIL-53(Fe)) nanocomposite photocatalysts were synthesized with PDA (PDA = polydopamine) and MIL-53(Fe) using a nature-mimicking method. The structures, morphologies, optical properties and thermal stabilities of all the synthesized materials were characterized using a series of methods. In particular, the separation efficiency of photogenerated charge significantly increased after the incorporation of PDA into MIL-53(Fe), which resulted in an elevated photocatalytic activity of PDA/MIL-53(Fe) compared with the control groups. The PDA/MIL-53(Fe) nanocomposite could accelerate the conversion of primary or secondary alcohols into the corresponding aldehydes or ketones with a high specificity by direct hole-involving oxidation under visible-light irradiation and room temperature. The catalysts could be cycled at least three times without a significant decrease in the catalytic activity and this result showed the excellent recyclability and stability of the catalysts.

In the present study, we reported the synthesis of a novel quinoline-based dendrimer-like ionic liquid. After characterization of the mentioned ionic liquid with suitable techniques such as Fourier transform infrared spectroscopy (FT-IR), energy dispersive X-ray spectroscopy (EDX), elemental mapping, thermogravimetric analysis (TGA) and derivative thermogravimetry (DTG), its catalytic performance was investigated in the synthesis of new pyridines with sulfonamide moiety via a cooperative vinylogous anomeric-based oxidation mechanism under mild reaction conditions. All target molecules were achieved in short reaction times and high yields.
