Intermediates in aromatic amino acid biosynthesis can serve as substrates for the synthesis of bioactive compounds. In this study we used two intermediates in the shikimate pathway of Escherichia coli, chorismate and anthranilate, to synthesize three bioactive compounds: 4-hydroxycoumarin (4-HC), 2,4-dihydroxyquinoline (DHQ), and 4-hydroxy-1-methyl-2(1H)-quinolone (NMQ). We introduced genes for the synthesis of salicylic acid from chorismate to supply the substrate for 4-HC and the gene encoding N-methyltransferase for the synthesis of N-methylanthranilate from anthranilate. Polyketide synthases and coenzyme (Co)A ligases were tested to determine the optimal combination of genes for the synthesis of each compound. We also tested several constructs and identified the best one for increasing levels of endogenous substrates for chorismate, anthranilate, and malonyl-CoA. With the use of these strategies, 255.4 mg/L 4-HC, 753.7 mg/L DHQ, and 17.5 mg/L NMQwere synthesized. This work provides a basis for the synthesis of diverse coumarin and quinoline derivatives with potential medical applications.

Enteroviruses (EV) are a group of positive-strand RNA ( + RNA) viruses that include many important human pathogens (e.g. poliovirus, coxsackievirus, echovirus, numbered enteroviruses and rhinoviruses). Fluoxetine was identified in drug repurposing screens as potent inhibitor of enterovirus B and enterovirus D replication. In this paper we are reporting the synthesis and the antiviral effect of a series of fluoxetine analogues. The results obtained offer a preliminary insight into the structure-activity relationship of its chemical scaffold and confirm the importance of the chiral configuration. We identified a racemic fluoxetine analogue, 2b, which showed a similar antiviral activity compared to (S)-fluoxetine. Investigating the stereochemistry of 2b revealed that the Senantiomer exerts potent antiviral activity and increased the antiviral spectrum compared to the racemic mixture of 2b. In line with the observed antiviral effect, the Senantiomer displayed a dose-dependent shift in the melting temperature in thermal shift assays, indicative for direct binding to the recombinant 2C protein.

Two magnetic nano catalysts of nickel and copper, Fe3O4@SiO2@DOP-BenPyr-M(II), (M=Ni and Cu) have been synthesized. These catalysts were applied as recoverable, efficient and new heterogeneous catalysts for the high yielding and room temperature one-pot procedure of selective oxidation of sulfides to sulfoxides and oxidative coupling of thiols to disulfides. In addition, the catalytic activity of Fe3O4@SiO2@DOP-BenPyr-Ni(II) was investigated as heterogeneous nanocatalyst for synthesis of 2,3-dihydroquinazolin-4(1H)-ones, 5-substituted 1H-tetrazoles and polyhydroquinolines. The synthesized catalysts were characterized by FT-IR, TGA, XRD, VSM, EDX, ICP and SEM techniques. Calcium dependent protein kinase 1 (CDPK1) is an essential Ser/Thr kinase that controls invasion and egress by the protozoan parasite Toxoplasma gondii. The Gly gatekeeper of CDPK1 makes it exquisitely sensitive to inhibition by small molecule 1H-pyrazolo[3,4-d]pyrimidine-4-amine (PP) compounds that are bulky ATP mimetics. Here we rationally designed, synthesized, and tested a series of novel PP analogs that were evaluated for inhibition of CDPK1 enzyme activity in vitro and parasite growth in cell culture. Optimal substitution on the PP scaffold included 2-pyridyl ethers directed into the hydrophobic pocket and small carbocyclic rings accessing the ribose-binding pocket. Further optimization of the series led to identification of the lead compound 3a that displayed excellent potency, selectivity, safety profile, and efficacy in vivo. The results of these studies provide a foundation for further work to optimize CDPK1 inhibitors for the treatment of acute and chronic toxoplasmosis.

In this study, preparation, characterization and catalytic application of a novel core-shell structured magnetic Tiloaded phenylene-based nanoporous organosilica (Mag@Ti-NOS) nanocomposite are developed. The Mag@TiNOS was synthesized via cetyltrimethylammonium bromide (CTAB) directed co-condensation of tetramethoxysilane (TMOS), tetrabutyl orthotitanate (TBOT) and 1,4-bis(triethoxysilyl)benzene (BTEB) over Mag@ SiO2 under alkaline conditions. This nanocomposite was characterized by using FT-IR, EDX, PXRD, VSM, SEM, TEM, XPS, ICP and TGA techniques. These analyses showed a core-shell structure with high chemical and thermal stability for the designed material. The Mag@Ti-NOS nanocomposite was employed as an effective, powerful and recyclable catalyst in the synthesis of tetrahydrobenzo[b]pyrans in H2O at 50 degrees C under ultrasonic conditions. This catalyst was recovered and reused several times without significant decrease in efficiency and stability. The recovered catalyst was analyzed by FT-IR, PXRD and SEM to study its stability during reaction process.

A novel approach was achieved for growing citric acid towards polymer-like graphene quantum dots (PGQD) with high efficiency in the presence of sodium hydroxide as a base catalyst. This protocol is completely safe, simple, fast, and efficient by a bottom up strategy. Thermal treatment of a mixture containing citric acid with NaOH at 300 degrees C gave PGQD during 5 min. The reaction afforded a new heterogeneous catalyst, Pd/PdO/Fe3O4@PGQD, in the presence of Pd and Fe3O4. The magnetically recoverable catalyst showed high activity in the oxidation of alkylarenes and alcohols using H2O2 as a green oxidant at room temperature. Comparison of the results with previous reports showed the efficiency of the catalyst to have high turnover numbers in mild reaction conditions.

An efficient CAN-mediated C-C bond formation in the oxidative reactions of xanthenes with enaminones under mild reaction conditions are described. A variety of 9H-xanthene-9-ylidene aldehydes could be synthesized in up to 90% yield via one-pot manner with good functional group compatibility, which achieved the simultaneous construction of carbon-carbon double bond and carbon-oxygen double bond.

Nickel(ii)complexes of N-4-ligands have been synthesized and characterized as efficient catalysts for the hydroxylation of benzene using H2O2. All the complexes exhibited Ni2+ -> Ni3+ oxidation potentials of around 0.966-1.051 V vs. Ag/Ag+ in acetonitrile. One of the complexes has been structurally characterized and adopted an octahedral coordination geometry around the nickel(ii) center. The complexes catalysed direct benzene hydroxylation using H2O2 as an oxygen source and afforded phenol up to 41% with a turnover number (TON) of 820. This is unprecedentedly the highest catalytic efficiency achieved to date for benzene hydroxylation using 0.05 mol% catalyst loading and five equivalents of H2O2. The benzene hydroxylation reaction possibly proceeds via the key intermediate bis(mu-oxo)dinickel(iii) species, which was characterized by HR-MS, vibrational and electronic spectral methods, for almost all complexes. The formation constant of the key intermediate was calculated to be 5.61-9.41 x 10(-2) s(-1) by following the appearance of an oxo-to-Ni(iii) LMCT band at around 406-413 nm. The intermediates are found to be very short-lived (t(1/2), 73-123 s). The geometry of one of the catalytically active intermediates was optimized by DFT and its spectral properties were calculated by TD-DFT calculations, which are comparable to experimental spectral data. The kinetic isotope effect (KIE) values (0.98-1.05) support the involvement of nickel-bound oxygen species as an intermediate. The isotope-labeling experiments using (H2O2)-O-18 showed 92.46% incorporation of O-18, revealing that H2O2 is the key oxygen supplier to form phenol. The catalytic efficiencies of complexes are strongly influenced by the geometrical configuration of intermediates, and stereoelectronic and steric properties, which are fine-tuned by the ligand architecture.

A new efficient approach to the synthesis of 6-alkenyl substituted pyridoxine derivatives has been developed. In vitro cytotoxicity of the obtained compounds against MCF-7 (ER+) breast cancer tumor cells was studied using the MTT assay. The most active compounds with IC50, MCF-7 < 10 mu M were also tested for cytotoxicity in vitro against MDA-MB-231 (ER-) breast adenocarcinoma cells and conditionally normal human skin fibroblasts (HSF). The patterns of structure-antitumor activity relationships of the obtained compounds were analyzed. The most active compounds were found to contain a six-membered ketal ring, a methyl group in position 5, a 3,4-dimethoxystyryl fragment in positions 2 or 6 of the pyridoxine ring, and a trans-configuration of the double bond. Using the most active compound 5a as a representative cytotoxic agent, we have demonstrated that it has high specificity and antiproliferative activity against MCF-7 (ER+) tumor cells (IC50 < 5 mu M), and a higher therapeutic index compared to the reference compound raloxifene (48 versus 5.8). Compound 5a decreased the mitochondrial membrane potential and increased the level of reactive oxygen species in MCF-7 cells, but not MDA-MB-231 cells. Compound 5a did not affect the distribution of cell cycle phases and induced apoptosis in MCF-7 cells, but not MDA-MB-231. Unlike compound 5a, raloxifene decreased mitochondrial potential, increased the ROS level, and induced apoptosis in both MCF-7 and MDA-MB-231 cells, which indicated a lack of selectivity for cells with estrogen receptor expression. It was also shown that compound 5a reduced the level of ER alpha expression in cells to a lesser extent than raloxifene and, unlike the latter, did not activate the PI3K/Akt signaling pathway.
