17beta-Hydroxysteroid dehydrogenase type 10 (17 beta-HSD10) is the only mitochondrial member of 17 beta-HSD family. This enzyme can oxidize estradiol (E2) into estrone (E1), thus reducing concentration of this neuroprotective steroid. Since 17 beta-HSD10 possesses properties that suggest a possible role in Alzheimer’s disease, its inhibition appears to be a therapeutic strategy. After we identified the androsterone (ADT) derivative 1 as a first steroidal inhibitor of 17 beta-HSD10, new analogs were synthesized to increase the metabolic stability, to improve the selectivity of inhibition over 17 beta-HSD3 and to optimize the inhibitory potency. From six D-ring derivatives of 1 (17-C = O), two compounds (17 beta-H/17 alpha-OH and 17 beta-OH/17aC CH) were more metabolically stable and did not inhibit the 17 beta-HSD3. Moreover, solid phase synthesis was used to extend the molecular diversity on the 3b-piperazinylmethyl group of the steroid base core. Eight over 120 new derivatives were more potent inhibitors than 1 for the transformation of E2 to E1, with the 4-(4-trifluoromethyl-3-methoxybenzyl)piperazin-1-ylmethyl-ADT (D-3,7) being 16 times more potent (IC50 = 0.14 mM). Finally, D-ring modification of D-3,7 provided 17 beta-OH/17 alpha-C CH derivative 25 and 17 beta-H/17 alpha-OH derivative 26, which were more potent inhibitor than 1 (1.8 and 2.4 times, respectively). A simple and efficient photocatalytic method for the synthesis of dihydropyrimidinones (DHPMs) and their derivatives via porous ultrathin carbon nitride nanosheets (p-CNNs) without solvents was demonstrated. The yields of 3,4-dihydropyrimidin-2(1H)-ones/thiones and their derivatives were up to 97.0%. Furthermore, the yield of 5-ethoxycarbonyl-6-methyl-4-phenyl-3,4-dihydropyrimidin-2(1H)-one in the 10th cycle retained 98.9% of its 1st value. Considering the environmental and economic benefits, this work exhibits various merits including excellent yields, environmental friendliness, inexpensiveness, and avoiding the use of solvents and metal-based photocatalysts. In addition, the excellent performance of the p-CNNs and environmentally benign reaction system have also been checked by the photocatalytic synthesis of 12-phenyl-9,9-dimethyl-8,9,10,12-tetrahydrobenzo[a]xanthen-11-one and 5-phenyl-1(4-methoxyphenyl)-3[(4-methoxy-phenyl)-aminol]-1H-pyrrol-2(5H)-one. This work paves a new way for carrying out a three-component reaction using metal-free photocatalysts under mild reaction conditions.

We have studied the generation of terahertz (THz) radiation via the interaction of an intense femtosecond laser pulse with aluminum (Al) and plastic targets pre-ablated by a picosecond ablation pulse. The measurements of the plasma shadowgraph and 1D-MULTI simulations show a similar micrometer density scale length of the pre-ablated plasma for the two types of targets after ablation. It is found that for the Al target, the intensity of THz radiation increased obviously under the condition of ablation, and only with the appropriate ablation delay time, the cutoff energy and charge of the accelerated protons also increased. 2D-simulations using the EPOCH program indicate that the strong enhancement of THz for the Al target is due to the increased number of energetic electrons from the interaction between the main laser and the ablated plasma. However, for the plastic targets, the intensity of THz radiation was almost vanished in the case of ablation, but the accelerated protons still existed and the cutoff energy even tended to increase. However, due to the lack of ability to simulate the molecular structure, we cannot distinguish between Al and plastic targets in the simulation. The mechanism of radiation suppression by ablation of the plastic target needs further investigation.

Benzodiazepines derivatives are nitrogen heterocyclic compounds that have various industrial, synthetic, and medicinal applications. Therefore, its potential fully justifies every effort towards the improvement of new, selective, and competent production of these heterocyclic compounds. The novelty of this study encompasses the synthesis of new hydroxylated analogs of 1,5-benzodiazepine together with the evaluation of their biological potential as antioxidants and antifungals candidates. Additionally, the reported antifungal activity against fungi of the Sporothrix schenckii complex inaugurates an unprecedented property for this class of compounds. The results of biological activities assays suggest that the novel 2,7-dimethyl-2,4-bis(4-hydroxyphenyl)-2,3-dihydro1H-1,5-benzodiazepine possesses a high antifungal activity against all fungal strains evaluated of the Sporothrix genus and outstanding total antioxidant capacity. Besides, in silico studies have evidenced that the hydroxylated derivatives have the best aqueous solubility and cell permeability profile, which makes them superior in comparison with the commercial antifungal itraconazole, a drug commonly used in the treatment of sporotrichosis.

Various aryl nitriles were readily synthesized from aerobic oxidation of substituted benzyl alcohols in deep eutectic solvent composed of choline chloride (ChCl) andp-toluenesulfonic acid (p-TsOH) in the presence of 2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPO), followed by condensation with hydroxylamine hydrochloride. High yields of corresponding nitriles have been obtained under mild reactions conditions. This strategy belongs to a novel and environmentally benign transition-metal-free one-pot cascade process for the synthesis of nitriles.

The synthesis and characterization of air-stable cationic mono nitrosonium Fe(I) PNP pincer complexes of the type [Fe(PNP)(NO)Cl](+) are described. These complexes are obtained via direct nitroslyation of [Fe(PNP)Cl-2] with nitric oxide at ambient pressure. On the basis of magnetic and EPR measurements as well as DFT calculations, these compounds were found to adopt a low-spin d(7) configuration and feature a nearly linear bound NO ligand suggesting (FeNO+)-N-I rather than (FeNO center dot)-N-II character. In response to the increasing interest of western consumers in high antioxidant activity of green tea but their low acceptance of its green odor, we employed a new starter culture, Wolfiporia cocos to tune flavor of green tea infusion. After submerged fermentation for 17 h, W. cocos changed the characteristic green odor to an attractive floral, jasmine-like, and slightly citrus-like flavor while preserving most of in vitro antioxidant activity. By application of mSBSE-GC-MS-O combined with sensorial tests, the formed pleasant aroma was mainly attributed to methyl anthranilate (OAV 802), linalool (OAV 190), 2-phenylethanol (OAV 165), and geraniol (OAV 118). Concurrently, the catechin profile determined by UHPLC-MS showed diverse reduction rates (10-50%) for the individual catechins after fermentation. Nevertheless, up to 80% of in vitro antioxidant activity in DPPH assay was preserved. Overall, our findings provide an innovative approach to naturally flavor green tea while retaining the antioxidant activity.

In the current research, two mesoporous structures of sodium zincate [Na2Zn(OH)(4)], spheres and aggregated particles, has been synthesized from sulfonation of melamine-formaldehyde (MF) and then bounding [Na2Zn(OH)(4)] particles onto the surface of sulfonated melamine-formaldehyde (SMF). Furthermore, in the present synthetic methodology the zinc oxide (ZnO) thin films on sulfonated melamine-formaldehyde was formed via the chemical deposition, and thermal treatment techniques of the sodium zincate. Several spectroscopic and analytical methods proved the structural and morphological properties of sodium zincate mesoporous structures based on sulfonated melamine formaldehyde [ SMF/Na2Zn(OH)(4)]. The experimental studies showed that these catalytic systems showed high activity in the Biginelli reaction for the synthesis of 3,4-dihydropyrimidin-2(1H)-one derivatives and will find recoverable potential application. The obtained results indicated that the synthesized mesoporous structures had a thermal stability near 300 degrees C, particle-size distribution around of 10-120 nm for spheres and the surface area of 6 and 2 m(2)/g for spheres and aggregated particles, respectively. We report herein the synthesis of a polymeric photosensitizer and its application in aerobic photooxidative hydroxylation of boronic acids. The polymeric photosensitizer was synthesized by the condensation of anthraquinone-2-carbonyl chloride (AQ-2-COCl) with poly (2-hydroxyethyl methacrylate) (PHEMA). The photo-oxidative hydroxylation of boronic acids using anthraquinone-containing-poly (2-hydroxyethyl methacrylate) (AQ-PHEMA) was then explored and shown to exhibit high efficiency and broad scope. Moreover, AQ-PHEMA could be easily recovered and reused for more than 20 times without significant loss of the catalytic activity.
