Novel 1-(1-benzoylpiperidin-4-yl)methanamine derivatives with high affinity and selectivity for serotonin 5-HT1A receptors were obtained and tested in four functional assays: ERK1/2 phosphorylation, adenylyl cyclase inhibition, calcium mobilization, and beta-arrestin recruitment. Compounds 44 and 56 (2-methylaminophenoxyethyl and 2-(1H-indol-4-yloxy)ethyl derivatives, respectively) were selected as biased agonists with highly differential signaling fingerprints that translated into distinct in vivo profiles. In vitro, 44 showed biased agonism for ERK1/2 phosphorylation and, in vivo, it preferentially exerted an antidepressant-like effect in the Porsolt forced swimming test in rats. In contrast, compound 56 exhibited a first-in-class profile: it preferentially and potently activated beta-arrestin recruitment in vitro and potently elicited lower lip retraction in vivo, a component of serotonergic syndrome. Both compounds showed promising developability properties. The presented 5-HT1A receptor-biased agonists, preferentially targeting various signaling pathways, have the potential to become drug candidates for distinct central nervous system pathologies and possessing accentuated therapeutic activity and reduced side effects.

Two copper(II) complexes with benzotriazole-based ligands having pyridyl and quinolinyl arms were synthesized in good yields and characterized fully via spectroscopic and crystallographic techniques. The crystal structures of the complexes revealed different coordination environments around copper(II) centers. The difference in structure was due to the steric effect of the ligands, which resulted in an octahedral and a square pyramidal geometry of the resulting complexes in the solid state. Both the air-stable complexes were utilized as precatalysts in catalytic click reactions and A(3) coupling.

5-Methylfurfural (MF) is a very useful chemical. Selective hydrogenation of biomass platform molecule 5-(hydroxymethyl)furfural (HMF) to MF using H-2 as the reducing agent is very attractive, but challenging because hydrogenation of C=O bond in HMF is more favourable than C-OH both kinetically and thermodynamically, and this route has not been realized. In this work, we prepare isolated single atomic catalysts (SACs) Pt-1/Nb2O5-Ov, Pd-1/Nb2O5-Ov, and Au-1/Nb2O5-Ov, in which single metal atoms are supported on oxygen defective Nb2O5 (Nb2O5-Ov). It is discovered that the SACs can efficiently catalyze the hydrogenation of HMF to MF using H-2 as the reducing agent with MF selectivity of >99% at complete conversion, while the selectivities of the metal nanocatalysts supported on Nb2O5 are very poor. A combination of experimental and density function theory (DFT) studies show that the unique features of the SACs for the reaction result from the cooperation of the Nb and Pt sites near the interface in the Pt-1/Nb2O5-Ov. The Pt atoms are responsible for the activation of H-2 and the Nb sites activate C-OH in the reaction. This work opens the way for producing MF by direct hydrogenation of biomass-derived HMF using H-2 as the reductant. Selective hydrogenation of 5-(hydroxymethyl)furfural (HMF) to 5-Methylfurfural using H2 as reductant is very attractive, but remains challenging. Here, the authors report that isolated single atomic catalysts can catalyze the reaction efficiently with selectivity >99% at complete conversion of HMF.

The direct amide synthesis from alcohols and amines applying various transition metal catalysts has been demonstrated as an attractive and promising process. Among various catalytic systems, N-heterocyclic carbene (NHC)-based ruthenium (Ru) ones have been testified to be active for this atom-economic transformation. Although a variety of imidazole-based NHC/Ru catalytic systems were reported to be active for this reaction, the benzimidazole-based analogs exhibited higher catalytic performance in most cases. However, these catalytic systems, which comprise a monodentate NHC ligand and a Cl or phosphine ligand as the key components, require relatively high catalyst loadings. In order to obtain more active and robust catalytic systems, we aim to bridge two monodentate ligands with one bidentate NHC ligand. Therefore, a number of CNHCC bidentate NHC precursors were designed and synthesized. Through screening of the NHC precursors and other reaction conditions, potent and phosphine-free bidentate NHC/Ru catalytic systems were discovered for the efficient amide synthesis. Interestingly, from the in situ generated catalytic system, two NHC/Ru intermediates were isolated and structurally confirmed by X-ray crystallography. Notably, these two complexes are active for the amide synthesis even at a low catalyst loading of 0.5 mol%, which could verify that they should be key intermediates during the catalysis. Probably, the current catalytic systems, featuring high efficiency and ready accessibility, could be valuable for more interesting applications.

The complex pathogenesis of Alzheimer’s disease (AD) has become a major obstacle in its therapy. One hopeful approach is to develop multifuntional ligands. Here, a total of 23 compounds based on nobiletin structure were designed and synthesized. The neuroprotective activities were evaluated for their antioxidant, anti-inflammatory and anti-A beta(42) (Amyloid beta-protein 42) neurotoxicity properties by MTT assays. The results demonstrated compound A12 was the most promising derivative against inflammatory (14.6 +/- 0.6 %) and cell damages induced by hydrogen peroxide (47.2 +/- 1.1 %), oxygen glucose deprivation (36.2 +/- 1.1 %) and A beta(42) (40.7 +/- 2.1 %). Molecular docking study revealed A12 formed a greater area of interactions with A beta(42) to prevent it from acquiring a beta sheet conformation for aggregation. Structure-activity relationships revealed the introduction of hydroxyl on ring B and aurone skeleton with a five-membered ring C could be benefit to increase their neuroprotective activities. In summary, A12 might represent a promising multifunctional neuroprotective agent against AD.

A novel catalytic system is reported for the construction of quinazolinones via the carbon-supported acid-catalyzed cascade coupling of isatoic anhydrides with amides and aldehydes. Synthesis of fluorescent molecules has drawn considerable attention due to their potential applications in life and material sciences. Here we report a metal-free approach to a new class of fluorophore, namely, 6-(2-hydroxyarypisoindolo[2,1-a]quinoxalin-11(5H)-ones exploiting ninhydrin ring system via base promoted tandem cyclization. The readily obtainable phenolic adducts of ninhydrin undergoes smooth reaction with o-phenylenediamine in refluxing ethylene glycol/Et3N via breaking of C-C bond, N-insertion, followed by heterocyclization to furnish the fully unsaturated isoindoloquinoxaline framework. Structures of the products are in good agreement with their IR, 1HNMR, (CNMR)-C-13, and mass spectral data. The photophysical properties of the synthesized compounds were studied by UV-vis absorption and fluorescence spectroscopic techniques under ambient conditions, and the results reveal an interesting property of large Stokes shift.
