Prostaglandins are among the most important natural isolates owing to their broad range of bioactivities and unique structures. However, current methods for the synthesis of prostaglandins suffer from low yields and lengthy steps. Here, we report a practicability-oriented synthetic strategy for the enantioselective and divergent synthesis of prostaglandins. In this approach, the multiply substituted five-membered rings in prostaglandins were constructed via the key enyne cycloisomerization with excellent selectivity (>20:1 d.r., 98% e.e.). The crucial chiral centre on the scaffold of the prostaglandins was installed using the asymmetric hydrogenation method (up to 98% yield and 98% e.e.). From our versatile common intermediates, a series of prostaglandins and related drugs could be produced in two steps, and fluprostenol could be prepared on a 20-gram scale.

Gastrodia elata is a famous traditional Chinese herb with medicinal and edible application. In this study, nine polybenzyls (1-9), including six new ones (2-5, 7 and 9), were isolated from the EtOAc extract of G. elata. Five compounds 1, 3, 4, 6 and 8 were found to activate melatonin receptors. Especially, compound 1 showed agonistic effects on MT1 and MT2 receptors with EC50 values of 237 and 244 mu M. For better understanding their structure-activity relationships (SARs), ten polybenzyl analogs were further synthesized and assayed for their activities on melatonin receptors. Preliminary SARs study suggested that two para-hydroxy groups were the key pharmacophore for maintaining activity. Molecular docking simulations verified that compound 1 could strongly interact with MT2 receptor by bonding to Phe 118, Gly 121, His 208, Try 294 and Ala 297 residues.

Old World (Africa) and New World (South America) arenaviruses are associated with human hemorrhagic fevers. Efforts to develop small molecule therapeutics have yielded several chemical series including the 4-acyl-1,6-dialkylpiperazin-2-ones. Herein, we describe an extensive exploration of this chemotype. In initial Phase I studies, R-1 and R-4 scanning libraries were assayed to identify potent substituents against Old World (Lassa) virus. In subsequent Phase II studies, R-6 substituents and iterative R-1, R-4 and R-6 substituent combinations were evaluated to obtain compounds with improved Lassa and New World (Machupo, Junin, and Tacaribe) arenavirus inhibitory activity, in vitro human liver microsome metabolic stability and aqueous solubility.

A series of 15 chalcones-bearing substituents at positions 2, 4, and 5 of rings A and B were synthesized using microwave-assisted Claissen-Smichdt condensation and evaluated for their activity against Giardia lamblia and Green monkey kidney cells. Five compounds exhibited activity against G. lamblia at IC50’s 3m exhibited the highest antigiardial activity (IC50 = 1.03 mu M), even more than the positive control (metronidazole, IC50 = 1.4 mu M), and selectivity (SI = 38.9). A preliminary SAR study suggested that electrophylicity has not relationship with antigiardiasic activity, and the docking study reveals that synthesized chalcones bind at zone 3 of colchicine site, therefore presumably the action mechanism of the synthesized chalcones does not follow the Michael acceptor mechanism.

The co-decomposition of non-noble metals into Ru nanoparticles (NPs) would provide multiple active centers as well as synergistically alter the reaction pathway, enhancing the catalytic hydrogenation performance. Herein, a facile route for synthesizing trielement Ru-Ni-Fe alloy NPs was proposed. The catalytic hydrogenation performance of NPs was measured using p-nitrophenol as a model. The synergistic effect of these three elements (Ru, Ni, and Fe) and synergistic catalysis of multiple crystal faces greatly improved the catalytic hydrogenation performance of Ru44Ni28Fe28 alloy NPs. Ru with more vacant orbitals showed a strong coordination with BF4- for the generation of active H species. Ni played a major role in transporting electrons and active H species, increasing the accessibility of catalytically active sites. Fe could cooperate with BH4- to produce active H species and promote electrons transfer. Ru44Ni28Fe28 alloy NPs could be reused and applied for the fabrication of films at the oil-water (ethyl acetate-water) interface. The densely packed Ru44Ni28Fe28 NP films were good Raman substrates for monitoring the complete conversion of 4-nitrothiophenol into 4-aminothiophenol. The rational design of Ru44Ni28Fe28 will broaden the application range of Ru-based catalysts and provide new insights into the rational design of other multisite alloy catalysts.

In this research, in order to synthesize a series of ethyl 2-amino-4-benzoyl-5-oxo-5,6-dihydro-4H-pyrano[3,2-c]quinoline-3-carboxylates, a green and an efficient method is proposed through one-pot three-component reaction of substituted arylglyoxals, ethyl cyanoacetate, and 4-hydroxyquinolin-2(1H)-one in the presence of terapropylammonium bromide as a catalyst in good yields. All synthesized new substances were characterized by FTIR, H-1-NMR, and C-13-NMR spectral data and elemental analysis.

We report a copper-catalyzed alkoxycyclization of tryptamine derivatives with O-2 as the sole oxidant, leading to a variety of C3a-alkoxypyrroloindolines in good yields with high diastereoselectivities. This reaction involves an interesting double catalytic cycle in which copper-catalyzed carboamination cyclization is favored to form the C-3 radical pyrrolidinoindoline intermediate, then a copper-catalytic radical alkoxylation reaction proceeds smoothly.

In the present investigation, a collection of twenty two nerol derivatives, containing 1,2,3-triazolic appendages, was synthesized and screened in vitro for their cytotoxic activity against HL60, Nalm6, and Jurkat human leukemia cells as well as against B16F10 (melanoma cell line). In most cases, derivatives were able to reduce cell viability. The most potent compound (Z)-4-(((3,7-dimethylocta-2,6-dien-1-yl)oxy)methyl)-1-(4-(trifluoromethoxy)benzyl)1H-1,2,3 triazole showed antiproliferative activity against Jurkat cells and reduced B16F10 cell migration. Physicochemical properties of the compounds were calculated in order to evaluate their potential for drug development. Most of the evaluated physicochemical parameters seemed to be favorable for drug development. In addition, for a better understanding of the biological activity results, 3D quantitative structure-activity relationship (QSAR) studies were carried out. 3D-QSAR studies indicate that the anticancer activities observed for the cell lines HL60 and Jurkat may occur by a similar mechanism of action and the same was found for the Nalm6 and B16F10 cell lines.

Characterization of volatile off-odor compounds in thermal duck egg gels was investigated by gas chromatography-olfactometry-mass spectrometry (GC-O-MS), odor activity values (OAVs) and aroma recombination. The results demonstrated that the off-odor of thermal duck egg mainly came from egg yolk (EY), and content of aldehydes was the highest in EY. Anaerobic treatment in nitrogen contributed to decreased off-odor sensory scores and volatile components of the heated egg gels. Aerobic EY gels (treated in air) were determined to contain eight volatile compounds with OAVs greater than 1, such as hexanal, nonanal, benzaldehyde, (E)-2-octenal, (E)-2-nonenal, (E, E)-2,4-decadienal, 1-octen-3-ol and 2-pentylfuran. However, three key volatile components (nonanal, benzaldehyde and (E)-2-nonenal) with OAVs greater than 1 were found in anaerobic EY gels. Moreover, on the basis of results of recombination and omission experiments, hexanal, (E)-2-octenal, (E, E)2,4-decadienal, 1-octen-3-ol and 2-pentylfuran were identified as key off-odor components in thermal EY gels. Sensory evaluation of the recombination model with five aroma compounds scored 4.38 out of 5 points. The major aroma profile of the off-flavor from egg EY gels included strong fatty, nutty, and mushroom aromas.

Allium is a large monocotyledonous genus, with many species of high economic importance. Knowledge of the pollination biology and the chemical ecology of pollination in this genus is far from being complete. We studied flower visitors of some Allium species in their native habitat and how these interaction partners communicate by olfactory cues. Floral volatiles of five Mediterranean species were investigated by dynamic headspace and thermal desorption-gas chromatography/mass spectrometry. Floral visitors were observed and captured. The physiological activity of scent components in antennae of flower visitors and congeneric species was tested by gas chromatographic/electroantennographic detections. In the scent samples of the five species, 36 volatile organic compounds (VOCs) were detected. The species-specific scent profiles were dominated either by one or two aromatic compounds, or by a monoterpene. We recorded several insects on the flowers/inflorescences of the different Allium species, mainly bees and flies, with only a few visitor taxa shared among the studied species. In the physiological measurements, specific/congeneric visitors did not only respond to VOCs of the species they visited, but also to VOCs of species on which they were not recorded. Our study shows that inflorescence scent and visitor patterns do not correlate, and that although single visitors detected compounds of various Allium species, there was only a limited overlap in visitor spectrum among the species. Our study also adds several compounds to the list of floral scents being EAD-active in bees and flies.
