Thiazole derivatives have been synthesized by the reaction of alpha-halo carbonyl compounds with thiosemicarbazones. Some pyridine derivatives have been synthesized by the reaction of chalcone with cyanothioacetamide and/or malononitrile. Spectroscopic methods have been used for elucidating molecular structures of the products. Cytotoxic activity of several derivatives has been tested against human breast cancer (MCF-7), human colon cancer (HCT-116) and human liver cancer (Hep-G2) by the SRB method. Most of compounds exhibit mild effect on the tested cell lines. One of thiazolidin-4-one derivatives has been characterized by moderate to strong effect on MCF-7 cell line.

BackgroundLaminitis is a common and serve disease which caused by inflammation and pathological changes of the laminar junction. However, the pathologic mechanism remains unclear. In this study we aimed to investigate changes of the gut microbiota and metabolomics in oligofructose-induced laminitis of horses.ResultsAnimals submitted to treatment with oligofructose had lower fecal pH but higher lactic acid, histamine, and Lipopolysaccharide (LPS) in serum. Meanwhile, oligofructose altered composition of the hindgut bacterial community, demonstrated by increasing relative abundance of Lactobacillus and Megasphaera. In addition, the metabolome analysis revealed that treatment with oligofructose decreased 84 metabolites while 53 metabolites increased, such as dihydrothymine, N3,N4-Dimethyl-L-arginine, 10E,12Z-Octadecadienoic acid, and asparagine. Pathway analysis revealed that aldosterone synthesis and secretion, regulation of lipolysis in adipocytes, steroid hormone biosynthesis, pyrimidine metabolism, biosynthesis of unsaturated fatty acids, and galactose metabolism were significantly different between healthy and laminitis horses. Furthermore, correlation analysis between gut microbiota and metabolites indicated that Lactobacillus and/or Megasphaera were positively associated with the dihydrothymine, N3,N4-Dimethyl-L-arginine, 10E,12Z-Octadecadienoic acid, and asparagine.ConclusionsThese results revealed that disturbance of gut microbiota and changes of metabolites were occurred during the development of equine laminitis, and these results may provide novel insights to detect biomarkers for a better understanding of the potential mechanism and prevention strategies for laminitis in horses.

Phenols are common precursors and core structures of a variety of industrial chemicals ranging from pharmaceuticals to polymers. However, the synthesis of site-specifically substituted phenols is challenging, and thus the development of new methods for this purpose would be highly desirable. Reported here is a protocol for palladium-catalyzed ortho-selective alkylation reactions of phenols with primary alcohols by a dearomatization-rearomatization strategy, with water as the sole by-product. Various substituted phenols and primary alcohols were compatible with the standard reaction conditions. The detailed mechanism of this transformation was also investigated.

Citronellol is one of the most widely used fragrances for bouquetting purposes and it is a starting material for synthesis of several other terpenoids. Nevertheless, few data have been reported on citronellol selective oxidation. Accordingly, we report our findings on the selective oxidation of citronellol with hydrogen peroxide using a set of titanosilicate catalysts with different morphologies and textural properties-conventional titanium silicalite 1 (TS-1), mesoporous TS-1, layered TS-1 and silica-titania pillared TS-1 and also studying the effect of the solvent used. Epoxidation of C6=C7 double bond was the main primary reaction in this system and trace signals of C5 allylic oxidation products were observed without formation of citronellal. Due to the presence of post-synthesis introduced additional Ti sites, the silica-titania pillared TS-1 (TS-1-PITi) provided the highest conversion among the tested catalysts; nevertheless, citronellol was oxidized over all the studied catalysts including conventional TS-1; therefore, showing that it penetrates even into MFI micropores (0.55 nm in diameter). When using acetonitrile as a solvent, the conversion was proportional to the titanium content of the catalyst. When studying the effect of the solvent, acetonitrile provided the highest epoxide selectivity (55%) while in methanol, 2-propanol and 1,4-dioxane, ring opening reactions caused epoxide decomposition.

Two new series of diethyl 2-[2-(substituted-2-oxo-1,2-dihydroquinolin-4-yl)hydrazono]-succinates 6a-g and 1-(2-oxo-1,2-dihydroquinolin-4-yl)-1H-pyrazoles 7a-f have been designed and synthesized. The structures of the synthesized compounds were proved by IR, mass, NMR (2D) spectra and elemental analyses. The target compounds were evaluated for their in vitro cytotoxic activity against 60 cancer cell lines according to NCI protocol. Consequently, seven compounds were further examined against the most sensitive cell lines, leukemia CCRFCEM, and MOLT-4. 5-Amino-1-(6-bromo-2-oxo-1,2-dihydroquinolin-4-yl)-1H-pyrazole-3,4-dicarbonitrile (7f) was the most active product, with IC50= 1.35 uM and 2.42 uM against MOLT-4 and CCRF-CEM, respectively. Also, it showed a remarkable inhibitory activity compared to erlotinib on the EGFR TK with IC50 = 247.14 nM and 208.42 nM, respectively. Cell cycle analysis of MOLT-4 cells treated with 7f showed cell cycle arrest at G2/M phase (supported by Caspases, BAX and Bcl-2 studies) with a significant pro-apoptotic activity as indicated by annexin V-FITC staining. Moreover, the docking study indicated that both the pyrazole moiety and the quinolin-2-one ring showed good fitting into EGFR (PDB code: 1M17). In order to interpret SAR of the designed compounds, and provide a basis for further optimization, molecular docking of the synthesized compounds to known EGFR inhibitors was performed. The study illustrated the effect of several factors on the compounds’ activity.

Iridium/Bronsted acid cooperative catalyzed asymmetric allylic substitution reactions at the C5 position of indolines have been reported for the first time. The highly efficient protocol allows rapid access to various C5-allylated products in good to high yields (48-97%) and enantioselectivities (82% to >99% ee) with wide functional group tolerance. This study evaluated the effect of different sugar smoking times on the flavour profiles of chicken drumsticks using an electronic nose (E-nose), electronic tongue (E-tongue), and headspace solid-phase microextraction gas chromatography-mass spectrometry (HS-SPME/GC-MS). The moisture content, water activity, pH, and L*-value decreased from 71.26% to 65.23%, 0.987 to 0.979, 6.66 to 5.36, and 61.84 to 52.34, respectively (P < 0.05). The a*-value and b*-value increased from 4.96 to 9.65 and 16.61 to 26.45, respectively (P < 0.05) with the increase in smoking times. Seventy-five volatile compounds were identified, and 18 volatile compounds were identified as key odour compounds and among them seven key volatile compounds with variable importance in projection (VIP) > 1 were detected. Principal component analysis of E-nose, E-tongue, and HS-SPME/GC-MS indicated that 3-, 4-, and 5-min smoking samples had similar odour and taste profiles. Sensory analysis indicated that the 4-min sample had increased overall acceptability. The correlation analysis of E-nose and key volatile compounds with odour activity value > 1 and VIP >1 confirmed that W1C, W3C, and W5C sensors were sensitive to aromatic compounds, and W2S sensor was sensitive to ketones. These results may provide guidance for smoked chicken producers to reasonably control flavour formation.

We disclosed an efficient Pd-catalyzed domino reaction for facile synthesis of spirooxindoles and [3,4]-fused oxindoles from alkene-tethered carbamoyl chlorides. This work aims to unravel the impact of disubstitution patterns on the physical properties and processing characteristics of indole-based aromatic polyesters. A series of hydroxyl-carboxylate (AB-type) monomers with 1,5-disubstituted indole and 3-6 methylene units was conveniently synthesized and used in bulk polycondensation to yield the corresponding polyesters with decent molecular weight. These new monomers and polyesters showed enhanced thermal stability compared to the previously reported monomers and polyesters with a 1,3-disubstituted indole structure. According to DSC results, these polyesters showed tunable glass transition temperatures (T-g similar to 57-80 degrees C), depending on the length of the aliphatic methylene units. DSC and WAXD measurements revealed that these polymers did not crystalize from melt, but the ones with 3 or 5 methylene units per repeating unit crystalized from solution. Finally, we demonstrated that the new polyesters with 1,5-disubstituted indole units could be crosslinked using sustainable aromatic aldehyde, which could further enhance their thermal properties.
