Exemplified for the Preparation of 6-chloroindole-3-carboxaldehyde (1) Phosphorus oxychloride (0.66 mL, 7 mmol) was added dropwise to anhydrous DMF (5 mL) at 0 C. under argon. A solution of 7-chloroindole (1 g, 6.6 mmol) in anhydrous DMF (15 mL) was added dropwise at room temperature and the resulting mixture was stirred for 2 h. The reaction mixture was poured into ice and saturated NaHCO3 and extracted with ethyl acetate. The combined organic solutions were washed with saturated NaCl (10 mL*3), dried over anhydrous MgSO4, filtered and concentrated to give 990 mg of product, 1, as a yellow-orange solid (83%).
