A series of seven luminescent copper(I) dithiophosphonate (dtp) clusters of the type Cu4L4 (L = S2PR(OR’)(-)) were formed from CuCl2 center dot 2H(2)O precursor in aqueous medium under ambient conditions. The dtp ligand serves the dual function of acting as a sacrificial reducing agent and cluster core stabilizer. The new clusters were characterized by H-1 and P-31 NMR and ESI-MS, and the single-crystal X-ray structures for two representative clusters [Cu-4{(S2P(1,4-C6H4OMe)(OR’)}4] (R’ = OCH(CH3)(2); CH2C6H5) were determined. The redox reaction yielded clusters in satisfactory yield, and all exhibited luminescence with lambda(em)(max) in the range 519-534 nm and half-lives in the range 10-14 mu s.

The use of covalent irreversible binding inhibitors is an established concept for drug development. Usually, the discovery of new irreversible kinase inhibitors occurs serendipitously, showing that efficient rational approaches for the rapid discovery of new drugs are needed. Herein, we report a virtual screening strategy that led to the discovery of irreversible inhibitors of FMS-like tyrosine kinase 3 (FLT3) involved in the pathogenesis of acute myeloid leukemia. A virtual screening library was designed to target the highly conserved Cys828 residue preceding the DFG motif by modification of reported reversible inhibitors with chemically reactive groups. Prospective covalent docking allowed the identification of two lead series, resulting in a massive increase in inhibition of kinase activity and cell viability by irreversible inhibitors compared to the corresponding reversible scaffolds. Lead compound 4b (BSc5371) displays superior cytotoxicity in FLT3-dependent cell lines to compounds in recent clinical trials and overcomes drug-resistant mutations.

The butyric acid scaffold is the base structure of several human metabolites that serve diverse and prominent biochemical roles including as oxidative sources of cellular energy and as substrates for biosynthesis. Derivatization of metabolites through incorporation of fluorine often alters bioactivity and can facilitate detection and analysis by nuclear magnetic resonance or positron emission tomography depending upon the fluorine isotope employed. We describe the synthesis of two new fluorinated butyric acids (and three related esters) that are derivatives of the metabolites acetoacetic acid and 3-hydroxybutyric acid. 4-Fluoro-3-hydroxybutyric acid is prepared from epoxy ester precursors via ring opening by triethylamine trihydrofluoride. 2-Fluoroacetoacetic acid is prepared by electrophilic fluorination of an acid-labile -keto ester. The gradual pH-dependent decarboxylation of 2-fluoroacetoacetic acid is investigated by (19) F NMR spectroscopy.

Headspace gas chromatography is frequently used for aroma profiling thanks to its ability to naturally exploit the volatility of aroma compounds, and also to provide chemical information on sample composition. Its main advantages rely on simplicity, no use of solvent, amenability to automation, and the cleanliness of the extract. In the present contribution, the most effective sampling (dynamic extraction), separation (multidimensional gas chromatography), and detection (mass spectrometry) techniques for untargeted analysis are exploited in combination, showing their potential in unraveling aroma profiles in fruit beers. To complete the overall analytical process, a neat workflow for data analysis is discussed and used for the successful characterization and identification of five different beer flavors (berries, cherry, banana, apple, and peach). From the technical viewpoint, the coupling of purge-and-trap, comprehensive two-dimensional gas chromatography, and mass spectrometry makes the global methodology unique, and it is for the first time discussed. Traditionally, conventional heat has been required for a large proportion of the oxidation and degradation process to utilise lignin from biomass. A photocatalysis system which is considered as a novel and green strategy for chemical reactions has been applied and a photo-MnO2 catalytic lignin oxidation method has been developed. In this study, we investigated a promising photocatalytic heterogeneous system for lignin oxidation. Recyclable MnO2 which is readily available and blue light which is a hazardless light are used in this system. 1-Phenylethanol was used as a model compound to study the suitable conditions for this system. After optimizing the reaction conditions, organosolv lignin, kraft lignin, and alkali lignin were applied to this system and the successful oxidation of lignins and their further degradation were shown.

A highly efficient nitrogen-doped carbon black (NCB)-supported PdBi alloy nanocatalyst has been fabricated via a facile coreduction wet chemical approach. The optimal PdBi/NCB shows outstanding catalytic performance with broad substrate scope, good functional group tolerance towards direct oxidative esterification of alcohols under mild conditions in a heterogeneous catalytic system with air as the sole oxidant. A variety of benzylic and allylic alcohols were smoothly reacted with methanol and even with long-chain aliphatic alcohols, providing desired esters in good to excellent yields. Moreover, the as-prepared catalyst is easily recycled and can be reused at least five times without a significant loss of catalytic activity. Superior catalytic activity is mainly attributable to the unique structure of the catalyst, including synergetic electronic effect between Pd and Bi, as well as modulated surface character by acidification and N doping for better active components’ anchoring and dispersion, as well as reactants’ adsorption. This study provides a facial, practical, eco-friendly and efficient catalytic system for oxidative esterification of alcohols and shows promising prospect in industrial production.

Lignin, as a byproduct from the paper industry and biorefineries, has been used as a renewable alternative to the conventional catalyst for future industrial application. However, due to the complex structure of lignin, its application has been considerably restricted. Herein, three types of lignin (soda lignin, kraft lignin, and enzymatic hydrolysis lignin) were prepared to illuminate the real impact of lignin interunit linkages and types of hydroxyl groups on the catalytic activity of cycloaddition reaction. Among these lignin/cocatalyst systems, a soda lignin/KI system showed excellent catalytic performance for the cycloaddition reaction at a low KI loading of 0.33 mol % (TON = 273). Particularly, soda lignin with abundant beta-O-4′ linkages, higher aliphatic C-a-OH groups, and phenolic hydroxyl groups as well as enriched carboxylic-OH groups could activate epoxides more efficiently and furthermore accelerate the cycloaddition reaction. Moreover, the dimeric intermolecular hydrogen bonding was dominant in soda lignin, which could also account for the higher catalytic activity of cycloaddition reaction. This work highlights the application of the soda lignin in a cycloaddition reaction and opens a new way for the utilization of lignin.

A set of heteropolyacid-based ionic heterogeneous catalysts (PW@IL-GO(m)) have been synthesized by intercalated heteropolyanions in ionic liquid (IL)-functionalized graphene oxide (GO) via amino-protonation and/or anion-exchange reaction. This immobilization method offered improved dispersity, accessibility and stability of active sites. The amphipathic nature of IL-GO sheets and porosity of PW@IL-GO(m) are beneficial to reducing mass transfer limitation in a water/organic substrate two-phase system. Then, these obtained hybrids were employed as efficient and robust catalysts for the selective oxidation of alcohols into carbonyl compounds using hydrogen peroxide as an oxidant under organic solvent-free condition. As expected, PW@IL-GO(m) catalysts showed even much better activity than the corresponding homogeneous HPW (phosphotungstic acid) catalyst. Particularly, PW@IL-GO(1.5) gave a benzyl alcohol conversion of 94% and benzaldehyde chemoselectivity of 91%. Moreover, owing to the heterogeneous nature and strong cation-anion interactions between cationic supporter and PW anions, the as-prepared catalyst presented advantages of convenient recovery, simple preparation and steady reuse, further providing the industrial application potential in sustainable oxidation reactions.

The synthesis of the hexasaccharide fragment of landomycin A is reported. Using p-toluenesulfonyl chloride mediated dehydrative glycosylation, we constructed the deoxy-sugar linkages in a stereoselective fashion without the need for temporary prosthetic groups to control selectivity. Presented here is a highly efficient procedure for the selective hydrogenation of aldehydes in the presence of ketones and its application in equilibrium displacement in a one-enzyme procedure catalysed by.-transaminase. A palladium-based heterogeneous catalyst was chemically synthesized from K2PdCl4 and 1,1′-binaphthyl-2,2′-ylenediamine and found to be suitable for selective hydrogenation of aldehydes using H2 of 1 atm, with a wide spectrum of aldehydes being efficiently and selectively reduced to the corresponding alcohols with a selectivity of up to 99%. This catalyst and its developed procedure can be successfully applied in pushing the equilibrium to product formation during transamination in a.-transaminase-promoted one-enzyme procedure via hydrogenation of the co-product, and an array of chiral amines was afforded in good to excellent yields of up to 99% with ees of up to > 99%.
