Natural and synthetic naphthoquinones are known for a large number of biological activities. Lawsone (2-hydroxy-1, 4-naphthoquinone) is a simplest naturally occurring compound obtained from dried henna (Lawsonia inermis) leaves. In literature, some lawsone derivatives have been reported to exhibit anticancer activity. Hence, a clean and facile one-pot protocol was developed for the synthesis of new aminonaphthoquinones derived from lawsone by three-component Mannich reaction, at room temperature for potential anti-cancer application. Herein we present a small library of Mannich bases with different amines and aromatic aldehydes with moderate to high yield. Synthesized compounds were characterized using various spectroscopic techniques. The anticancer activity (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay) along with nuclear morphology assessment (4 ‘,6-diamidino-2-phenylindole or DAPI staining), apoptosis assessment (acridine orange/ ethidium bromide staining), hemolysis and DNA ladder assay evaluated on human liver carcinoma cell line HepG2 are presented.

These domino processes allow divergent syntheses of two significant N-heterocycles, indoles and quinolines, from the same substrate by tuning reaction parameters, which seems to invoke two distinct mechanisms. The nature of the ligand and alcoholic solvent had a profound influence on the selectivity and efficiency of these protocols. An amine-functionalized metal organic framework (MOF) was used as highly efficient and recyclable heterogeneous catalyst for Knoevenagel condensation of various aromatic aldehydes and ketones in ethanol. The catalytic efficiency was demonstrated by the high conversion of the reactants with 100% selectivity under microwave (mu W) irradiation, while dramatically reduced the reaction time as compared to the conventional heating method. Importantly, the MOF maintained its structural integrity after catalytic reactions and it could be reused several times without remarkable loss of activity.

We revealed a catalyst-free, green and rapid one-pot four-component tandem strategy for the preparation of dihydropyrano[2,3-c]pyrazoles – a biologically significant scaffold – via Knoevenagel-Michael cyclocondensation based on green chemistry principles. Highlights of the current practice are the application of non-hazardous reaction circumstances, catalyst-free, operational simplicity, use of inexpensive initiating substances, isolation of pure product via easy filtration thus preventing the requirement for column chromatography, metal-free, excellent yields, time-saving aspects of the reaction. However, the green ethylene glycol (E-G) can be recycled at least six times with no considerable reduction in activity making it greatly advantageous in addressing the industrial requirements and environmental worries.

Polymeric graphitic carbon nitride (g-C3N4) supported CuBr2 hybrid material (CuBr2@g-C3N4) was developed by impregnation of CuBr2 on g-C3N4 sheets. The morphology and physiochemical properties were analyzed by powder XRD, SEM, and HRTEM studies. The hybrid CuBr2@g-C3N4 material showed excellent selectivity in the aerobic oxidation of primary/secondary alcohols and benzoins to the corresponding carbonyl compounds in absence of any oxidizing agents. Further, CuBr2@g-C3N4 demonstrated direct oxidation of toluene derivatives to corresponding aldehydes in excellent yields. The g-C3N4 support provides excellent stability and heterogeneity to CuBr2 and was recovered simply by filtration and reused for ten times without appreciable loss in catalytic activity.

Leishmaniasis is a neglected parasitic disease, and current treatment includes limitations of toxicity, variable efficacy, high costs and inconvenient doses and treatment schedules. Therefore, new leishmanicidal drugs are still an unquestionable medical need. In this paper we described the design conception of a new framework, the carbamoyl-N-aryl-imine-urea, to obtain putative leishmanicidal drug-candidates. Compounds 9a-e and 10a-e were designed and synthesized and their leishmanicidal activity was studied in comparison to pentamidine, miltefosine and meglumine antimoniate. The conformational profile of the new carbamoyl-N-aryl-imine-urea framework was investigated by X-ray diffraction studies, using compound 9a as a model. The plasma stability of this putative peptide mimetic subunit was studied for compound 10e (LASSBio-1736). Among the congeneric series, LASSBio-1736 was identified as a new antileishmanial drug-candidate, displaying plasma stability, cytotoxicity against amastigote forms of L. amazonensis and L. braziliensis, and leishmanicidal activity in a cutaneous leishmaniasis murine model, without preliminary evidence of hepatic or renal toxicity.

Two magnetic nano catalysts of nickel and copper, Fe3O4@SiO2@DOP-BenPyr-M(II), (M=Ni and Cu) have been synthesized. These catalysts were applied as recoverable, efficient and new heterogeneous catalysts for the high yielding and room temperature one-pot procedure of selective oxidation of sulfides to sulfoxides and oxidative coupling of thiols to disulfides. In this work, the use of global models (able to describe the whole sample) is proposed as an alternative to the use of individual models for each solute. Global models contain parameters that are specific for each solute, while other parameters -related to the column and solvent- are common for all solutes. A special regression procedure is presented for the construction of global models, which are applied to predict highly complex chromatograms, such as chromatographic fingerprints, for diverse experimental conditions in isocratic and gradient elution. Another interesting application is the prediction of molecular properties, such as log P-o/w from the specific solute parameters of the global models. The examined adapted models are based on the equations proposed by Snyder, Schoenmakers, Neue and Kuss, Jandera, and Bosch Roses to describe the retention. In all cases, the predictive capability was very satisfactory. Two cases of study were considered: chromatograms of camomile extracts analysed using acetonitrile gradients, and a set of 145 known compounds in a wide range of structures and functionalities, eluted isocratically with acetonitrile/water mobile phases. Herein we report the generation of benzylic lithiums via an iodine-lithium exchange reaction on benzylic iodides performed in continuous flow using tBuLi as the exchange reagent. The resulting benzylic lithium species are trapped in situ by carbonyl electrophiles under Barbier conditions, resulting in benzylic secondary and tertiary alcohols. This flow procedure further allows the generation of highly reactive heterobenzylic lithium compounds, which are difficult to generate under batch conditions. A general scale-up was possible without further optimization.

This article describes a green method for regeneration of carbonyl compounds from various types of oxime compounds under microwave radiation using I-2 and aqueous saponin solution isolated from Shikakai. Effect of saponin concentration on yield percentage of regenerated different types of carbonyl compounds has been discussed. A correlation has been established between saponin concentration and yield percentage of carbonyl compounds. Mechanism of interaction between oxime and saponin is established on the basis of density functional theory. In addition, the quantum chemical parameters for saponin have been determined. Furthermore, electrostatic surface analysis of the saponin is carried out to confirm the mechanism of interaction between saponin and oximes. (C) 2020 Elsevier B.V. All rights reserved.
