The synthesis of 2-amino-4H-chromenes through one-pot multicomponent reactions has received great attention due to the high atom efficiency and the broad bioactivities of the products. Here, the catalytic performances of a series of functional ionic liquids towards this type of reaction were investigated and the results showed that the ionic liquid, choline acetate ([Choline][Ac]), could efficiently promote this reactions under room temperature without the necessary of organic solvents. The reaction is easy to be conducted and the following work-up procedures are very simple. The pure products can be obtained through extraction and washing procedures, no column separation procedures are needed. Intriguingly, this reaction system can be easily scaled up to multi-gram, suggesting its perspective in industrial applications. In addition, the ionic liquid [Choline][Ac] can be easily prepared from cheap and biocompatible materials, and it can be feasibly recycled and reutilized for at least five cycles. The oxidation of aldehydes is one of the most prevalent methods for the synthesis of a diverse range of carboxylic acids. We herein present a performic acid generator and its application for aldehyde oxidations under continuous flow conditions. Low molecular weight performic acid, an environmentally benign and inexpensive oxidant, was readily formed in situ from formic acid and hydrogen peroxide. The safety hazards typically encountered when manipulating this potentially explosive reagent were eliminated, while at the same time a clean and efficient access to valuable carboxylic acids was ensured. By taking advantage of concentrated or even neat aldehyde streams, the amount of organic solvents utilized, and thus waste formation, was successfully minimized as demonstrated by E-factors in the range of 5-18. The process proved applicable for a wide range of aromatic as well as aliphatic aldehydes, while its synthetic utility and stability were corroborated by scale-out experiments leading to multigram-scale syntheses.

Natural product (NP) structures are a rich source of inspiration for the discovery of new biologically relevant chemical matter. In natural product inspired pseudo-NPs, NP-derived fragments are combined de novo in unprecedented arrangements. Described here is the design and synthesis of a 155-member pyrroquinoline pseudo-NP collection in which fragments characteristic of the tetrahydroquinoline and pyrrolidine NP classes are combined with eight different connectivities and regioisomeric arrangements. Cheminformatic analysis and biological evaluation of the compound collection by means of phenotyping in the morphological cell painting assay followed by principal component analysis revealed that the pseudo-NP classes are chemically diverse and that bioactivity patterns differ markedly, and are dependent on connectivity and regioisomeric arrangement of the fragments.

A comprehensive approach of untargeted and targeted fatty acid bioprofiling of different royal jelly commercial and pharmaceutical products based on HPTLC-image analysis and melanoma cytotoxic activity together with chemometric analysis was applied in this study for discovering efficient biomarkers. Principal component analysis based on HPTLC-image analysis fingerprints of fatty acid loading plots were used to determine the chemical markers responsible for classification of royal jelly samples into fresh and lyophilized ones. These markers were identified using the HPTLC-MS technique as 8-hydroxyoctanoic acid, 3,10-dihydroxydecanoic acid, 10-hydroxy-2-decenoic acid, decanedioic acid and 10-hydroxydecanoic acid. These discriminating markers were quantified via the HPTLC-imaging technique for targeted profiling using two different methods: parametric and non-parametric regression. The non-parametric regression method exhibited superiority in terms of linearity, accuracy and precision. Biomarkers were determined from the 3D-loading plot of orthogonal projection to latent structures model based on the fatty acid quantitative data together with the melanoma cytotoxic activity data. 10-Hydroxy-2-decenoic acid showed the greatest reduction in melanoma cell viability followed by decanedioic acid then 8-hydroxyoctanoic acid. The present study is considered the first attempt to discriminate fresh and lyophilized royal jelly samples based on their holistic lipidomic profile to discover efficient fatty acid reducing melanoma cell viability.

A one-pot strategy for the straightforward Mn(III)-mediated phosphinoylation of aldehyde hydrazones with diphenylphosphine oxide to furnish alpha-iminophosphine oxides is described. This mild and practical method allows the direct use of aldehydes as substrates in one pot to generate the hydrazones, which are then engaged in situ by the phosphorus reagent in the presence of Mn(OAc)(3) oxidant. A simple and efficient synthesis of novel 6,6 ‘-(arylmethylene)bis(benzo[a]phenazin-5-ol) derivatives has been developed via a sequential one-pot, two-step, pseudo-five-component tandem reaction starting from 2-hydroxynaphthalene-1,4-dione, o-phenylenediamine, and aromatic aldehydes in the presence of 2-aminopyridine as co-catalyst and p-TsOH as catalyst at 90 degrees C under solvent-free conditions. A facile synthesis of dehydroepiandrosterones derived by C-16 spiro-pyrrolidine and their cytotoxic evaluation are reported. Seven derivatives, 3a-3g, were prepared by the [3 + 2] cycloaddition reaction of the 16-arylidene dehydroepiandrosterone and the azomethine ylide, where the azomethine ylide was generated in situ from 11H-indeno[1,2-b]quinoxalin-11-one and sarcosine. Their cytotoxicity was evaluated on brine shrimp assay and the bioactivities represented by LC50 values were found in the range of 6.19-27.2 mu g/mL. Among them, the activities of 3d and 3g are the best, as manifested by LC50 values less than 10 mu g/mL. All products were confirmed by NMR, HR MS and X-ray analysis.
