Cladosporine A (1), a new indole diterpenoid alkaloid, was isolated from the extract of a fungal strain Cladosporium sp. JNU17DTH12-9-01. Its structure was elucidated by extensive spectroscopic analysis, and the absolute configurations were determined by electronic circular dichroism (ECD) experiments. This is the first report of the presence of indole diterpenoid alkaloid in the genus Cladosporium. The antimicrobial activities against Staphylococcus aureus 209P, Escherichia coli ATCC0111, Aspergillus niger R330, and Candida albicans FIM709 were evaluated. Compound 1 showed MICs of 4 mu g/mL and 16 mu g/mL against S. aureus 209P and C. albicans FIM709, respectively. [GRAPHICS] .

Covalent or Non-Covalent? A Mechanistic Insight into the Enantioselective Bronsted Acid Catalyzed Dearomatization of Indoles with Allenamides

The reaction mechanism of the enantioselective Bronsted acid catalyzed dearomatization of C(2),C(3)-disubstituted indoles with allenamides is investigated by means of density functional theory (DFT) calculations and ESI-MS analysis. The first step of the process (rate-determining step) is the formation of a covalent adduct between allenamide and the chiral organo-promoter. The resulting chiral alpha-amino allylic phosphate undergoes dearomative condensation with indoles. In the first step, the indole moiety remains bonded to the catalyst through strong hydrogen contacts. It can take on two different orientations that make the Re or Si prochiral face available to the subsequent electrophilic attack of allenamide. The attack on the indole faces originates two reaction paths leading to different stereoisomeric products, which differ in the configuration of the new stereocenter at the C3-indole position.

Mn(OAc)(3)-Mediated Regioselective Radical Alkoxycarbonylation of Indoles, Pyrimidinones, and Pyridinones

Mn(OAc)(3)-mediated alkoxycarbonylation of indoles, pyrimidinones, and pyridinones with alkyl carbazates is reported. The reactions proceed through a radical process to afford regioselectively 3-carboxylated indoles, 5-carboxylated pyrimidinones, and 3-carboxylated pyridinones in moderate to good yields under mild reaction conditions.

Design, synthesis and structure activity relationships of indazole and indole derivatives as potent glucagon receptor antagonists

A novel series of indazole/indole derivatives were discovered as glucagon receptor (GCGR) antagonists through scaffold hopping based on two literature leads: MK-0893 and LY-2409021. Further structure-activity relationship (SAR) exploration and optimization led to the discovery of multiple potent GCGR antagonists with excellent pharmacokinetic properties in mice and rats, including low systemic clearance, long elimination half-life, and good oral bioavailability. These potent GCGR antagonists could be used for potential treatment of type II diabetes.
