Simultaneous chemosensing of tryptophan and the bacterial signal molecule indole by boron doped diamond electrode

A simple and robust chemosensing approach using a boron-doped diamond (BDD) electrode has been developed and applied to analyze tryptophan (TRP) and indole during the growth of Escherichia coli in a complex growth medium. The bacterial enzyme tryptophanase catalyzes TRP to indole, an emerging signaling molecule. The process can now be monitored using electrochemistry, in a method far beyond the traditional identification protocols. Electroanalysis in a non-aqueous medium comprising acetonitrile (ACN) and tetrabutylammonium hexafluorophosphate (TBAH) is capable of separating the oxidation peak of TRP from that of indole. Mechanisms are postulated for the electrochemical oxidation of indole and TRP in ACN chosen because of its wider potential range, proton acceptor property, and solubilization of analytes. The electrochemical oxidation of TRP involves the elimination of two electrons. With a detection limit of 0.5 mM for both indole and TRP, this chemosensing approach is sufficient to monitor the level of these two biomolecules during the bacterial growth period. Cu(II)-Catalyzed Oxidative Trifluoromethylation of Indoles with KF as the Base

This paper offers an efficient copper-catalyzed oxidative trifluoromethylation of indoles with low-cost CF3SO2Na via C-H activation. Notably, the use of a base is crucial for the trifluoromethylation of indoles. This reaction proceeds efficiently in good to excellent yields and is tolerance of a broad range of functional groups. Furthermore, melatonin, a medicine for sleep disorders, is converted to its 2-CF3 analogue in 68% yield. Studies of possible reaction pathways suggest that this reaction proceeds through a radical process.

ERF Gene Clusters: Working Together to Regulate Metabolism

Plants produce structurally diverse specialized metabolites, including bioactive alkaloids and terpenoids, in response to biotic and abiotic environmental stresses. The APETALA2/ETHYLENE RESPONSE FACTOR (AP2/ERF) family of transcription factors (TFs) play key roles in regulating biosynthesis of specialized metabolites. Increasing genomic and functional evidence shows that a subset of the ERF genes occurs in clusters on the chromosomes. These jasmonate-responsive ERF TF gene clusters control the biosynthesis of many important metabolites, from natural products, such as nicotine and steroidal glycoalkaloids (SGAs), to pharmaceuticals, such as artemisinin, vinblastine, and vincristine. Here, we review the function, regulation, and evolution of ERF clusters and highlight recent advances in understanding the distinct roles of clustered ERF genes and their possible application in metabolic engineering.

Facile Synthesis of Polyfunctional Indole-Pyrido[2,3-d]Pyrimidine Hybrids Using Nickel-Incorporated Fluorapatite Encapsulated Iron Oxide Nanocatalyst and Study of Their Antibacterial Activities

Fe3O4@FAp@Ni was synthesized and has been used as a new and efficient recyclable magnetic nanocatalyst for the synthesis of polyfunctional indole-pyrido[2,3-d]pyrimidine hybrids. The Fe3O4@FAp@Ni was conveniently separated by an external magnet and could be reused at least six times without any appreciable change in catalytic activity. This novel protocol furnished the products in high to excellent yield (75-95%) and short reaction time. The antibacterial properties of the products were also evaluated and some of them exhibited promising activities.
