This paper presents a comparative study of endophytic bacteria from cultivated (Oryza sativa) and wild rice (Oryza rufipogon) plants and their functional traits related to plant growth promotion. A total of 70 bacterial isolates were characterized by both biochemical and molecular identification methods. Taxonomic classification showed dominance of three major phyla, viz, Firmicutes (57.1%), Actinobacteria (20.0%) and Proteobacteria (22.8%). Screening for in vitro plant growth-promoting activities revealed a hitherto unreported endophytic bacterium from wild rice germplasm, Microbacterium laevaniformans RS0111 with highest indole acetic acid (28.39 +/- 1.39 mu g/ml) and gibberellic acid (67.23 +/- 1.83 mu g/ml) producing efficiency. Few other endophytic isolates from cultivated rice germplasm such as Bacillus tequilensis RHS01 showed highest phosphate solubilizing activity (81.70 +/- 1.98 mu g/ml), while Microbacterium testaceum MKLS01 and Microbacterium enclense MI03 L05 showed highest potassium (53.42 +/- 0.75 mu g/ml) and zinc solubilizing activity (157.50%). Fictibacillus aquaticus LP20 05 produced highest siderophore (64.8%). In vivo evaluation of plant growth-promoting efficiencies of the isolates showed that Microbacterium laevaniformans RS0111, Microbacterium testaceum MKLS01 and Bacillus tequilensis RHS 01 could increase rice grain yield by 3.4-fold when compared to the control group. This study indicates the potentiality of rice endophytes isolates as an effective bioinoculants.

Physiological impacts of zero valent iron, Fe3O4 and Fe2O3 nanoparticles in rice plants and their potential as Fe fertilizers

Fe-based nanoparticles (Fe-based NPs) have great potential as a substitute for traditional Fe-fertilizer; however, their environmental risk and impact on plant growth are not fully understood. In this study, we compared the physiological impacts of three different Fe-based NP formulations: zero-valent iron (ZVI), Fe3O4 and Fe2O3 NPs, on hydroponic rice after root exposure for 2 weeks. Fe-normal (Fe(+)) and Fe-deficiency (Fe(-)) conditions were compared. Results showed that low dose (50 mg L-1) of ZVI and Fe3O4 NPs improved the rice growth under Fe( -) condition, while Fe2O3 NPs did not improve plant growth and caused phytotoxicity at high concentration (500 mg L-1). Under Fe(+) conditions, none of the Fe-based NPs exhibited positive effects on the rice plants with plant growth actually being inhibited at 500 mg Cl evidenced by reduced root volume and leaf biomass and enhanced oxidative stress in plant. Under Fe(-) condition, low dose (50 mg L-1) of ZVI NPs and Fe3O4 NPs increased the chlorophyll content by 30.7% and 26.9%, respectively. They also alleviated plant stress demonstrated by the reduced oxidative stress and decreased concentrations of stress related phytohormones such as gibberellin and indole-3-acetic acid. Low dose of ZVI and Fe3O4 NPs treatments resulted in higher Fe accumulation in plants compared to Fe2O3 NPs treatment, by down-regulating the expression of IRT1 and YSL15. This study provides significant insights into the physiological impacts of Fe-based NPs in rice plants and their potential application in agriculture. ZVI and Fe3O4 NPs can be used as Fe-fertilizers to improve rice growth under Fe-deficient condition, which exist in many rice-growing regions of the world. However, dose should be carefully chosen as high dose (500 mg Cl in this study) of the Fe-based NPs can impair rice growth. (C) 2020 Elsevier Ltd. All rights reserved.

Impacts of dietary supplementation with p-coumaric acid and indole-3-acetic acid on survival and biochemical response of honey bees treated with tau-fluvalinate

Pollinator populations are in decline worldwide. Multiple factors have been cited as potential causes to these declines. In honey bees, a combination of stressors is known to cause colony losses. Adequate nutrition is a key factor for honey bee growth and colony development. Several studies show that the nutritional quality of the diet is directly proportional to the ability of the bee to face challenges or stressors. We explored the effect of p-coumaric (600 mu M) and indole-3-acetic acid (2, 20 or 200 mu M) supplementation on the survival and activity of key detoxification enzymes of honey bees exposed to tau-fluvalinate. The dietary supplementation with p-coumaric and indole-3-acetic acids (20 mu M) enhanced the survival of bees exposed to tau-fluvalinate (approximately 20%). We also showed that dietary p-coumaric acid increased the levels of cytochrome P450 and glutathione reductase activity in bees treated with tau-fluvalinate, as well as in the untreated controls, while glutathione-S-transferase activity was lower in treated bees than in untreated. In bees fed with indole-3-acetic acid, cytochrome P450 showed increased levels, however, glutathione-S-transferase showed the lowest activity. Moreover, the results showed that supplementation with p-coumaric and indole-3-acetic acids did not alter acetyl cholinesterase activity, nor did treatment with tau-fluvalinate. Altogether, the enzymatic changes related to the detoxification mechanisms observed in bees that were fed with p-coumaric and indole-3-acetic acids could be responsible for the increased survival of bees treated with tau-fluvalinate compared to those that received a control diet. The results presented in this study, together with previous studies, provide evidence of the importance of dietary phytochemicals in the response of honey bees to pesticide exposure. Moreover, these results are the first report of the beneficial effect of the phytohormone indole-3-acetic acid on the survival of honey bees treated with tau-fluvalinate.
