Electron-rich aza-aromatic compounds such as indoles and pyrroles are structures of particular interest and importance in organic chemistry. A useful methodology for the regioselective introduction of the sulfenyl group into electron-rich aza-aromatics using S-alkyl- and S-arylthiophthalimides as sulfenylating agents is described. Catalytic amounts of CeCl3· 7H2O/NaI are crucial to the promotion of this regioselective carbon-sulfur-bond-forming electrophilic aromatic substitution reaction. The reaction occurred under mild conditions, and the products were obtained in good to excellent yields. The method represents an efficient preparation of sulfenyl aza-aromatics, which are useful intermediates for important organic transformations, due to the great importance of functionalized indoles among natural compounds and pharmaceutical products. Friedel-Crafts reaction of indoles with alpha-amido sulfones in the presence of montmorillonite K-10 leads unexpectedly to 3-(1-arylsulfonylalkyl) indoles in good yield. The obtained products can be further desulfonylated under reductive or alkylative conditions giving linear and branched 3-substituted indoles.

Resistance developments against established antibiotics are an emerging problem for antibacterial therapies. Infections with Staphylococcus aureus and methicillin-resistant S. aureus (MRSA) have become more difficult to treat with standard antibiotics that often fail, especially against MRSA. In consequence, novel antibiotics are urgently needed. Antibiotics from natural sources own complicated structures that cause difficulties for a chemical synthetic production. We developed novel small-molecule antibacterials that are easily accessible in a simple one-pot synthesis. The central indolonaphthalene core is substituted with indole residues at various positions. Both the varied indole substitutions and their positions at the molecular scaffold influence the determined antibacterial activity against the evaluated Staphylococcus strains. Best activities have been found for 5-chloro,-cyano, and-hydroxyl indole substitutions. Therefore, first promising lead compounds could be identified that are nontoxic in human HEK and SH-SY5Y cells and exceed the activity of used standard antibiotics, especially against MRSA.

A novel TEMPO/CoCl2-promoted aerobic oxidation of indoles was developed. The reaction provided one-step access to tryptanthrin derivatives in moderate to good yields and excellent regioselectivity via a cascade process. The reactions could be carried out under mild reaction conditions with varying functional group tolerance, especially halogen functional groups. Mechanistic studies disclosed that the oxygen atom in the desired product originated from molecular dioxygen.

A convenient method for the synthesis of 3-methylthioindoles has been established which does not use smelly compounds such as thiol derivatives. The method, which introduces an alkyl- or arylthio-group into the C3-position of the indole skeleton, was extended to the direct introduction of a methylthio or bromo group at the C2-position using 3-methylthioindoles. No dimerization occurred, and the reaction mechanism was confirmed. The products have the partial structure of potent anti-methicillin-resistant Staphylococcus aureus (anti-MRSA) bromomethylthioindoles (MC 5-8) isolated from marine algae. Furthermore, this reaction could be applied to the synthesis of 3,3-diindolyl thioether which is a core structure of Echinosulfone A.

In situ spectroelectrochemical studies of the fluorescence of 5- substituted indole trimer films

A novel in situ spectroelectrochemical cell has been constructed for the simultaneous measurement of fluorescence and current-voltage characteristics during redox cycling. This involves the use of a detachable rotating disc electrode (RDE), which allows the characterisation of luminescent redox active films produced electrochemically under controlled hydrodynamic conditions. Using this cell, the fluorescence of 5-cyanoindole (CI) and indole-5-carboxylic acid (ICA) films has been measured as a function of redox composition during cyclic voltammetry at slow sweep rates. At faster sweep rates hysteresis has been observed in the recorded fluorescence-charge response, indicative of a structural change in the film. The rate of this structural change appears to be decreased by linking the trimer redox centres in the film and by switching from CI to ICA, consistent with the influence of intertrimer hydrogen bonding. Variation in the observed fluorescence emission as a function of wavelength has also been observed, which gives spatial information on the nature of the film redox reaction. This technique shows potential as a method for probing the kinetics and mechanisms of the reaction of electrochemiluminescent films.

Organocatalytic synthesis of alpha-quaternary amino acid derivatives via aza-Friedel-Crafts alkylation of indoles with simple alpha-amidoacrylates

The first (organo)catalytic method for regio- and chemoselective aza-Friedel-Crafts (FC) alkylation of indoles and pyrroles with commercially available methyl alpha-acetamidoacrylates has been discovered. It minimizes/eliminates common competing reactions that occur due to the high and multiatom-nucleophilic character of indole and pyrrole. Diverse quaternary alpha-amino acids were successfully prepared in good yield and high selectivity using low catalyst loading. The enantioselective variant using BINOL-derived phosphoric acids was also explored with indole providing the desired F-C alkylation product with moderate enantioselectivities.
