In the current work, we show that glycyrrhizin, a muscle relaxant used in Traditional Chinese Medicine, can activate oxidative burst and actin remodelling in tobacco BY-2 cells, which could be suppressed by diphenylene iodonium, an inhibitor of NADPH oxidases. Glycyrrhizin caused a dose-dependent delay of proliferation, and induced cell death, which was suppressed by addition of indole-acetic acid, a natural auxin that can mitigate RboH dependent actin remodelling. To test, whether the actin remodelling induced by glycyrrhizin was followed by activation of defence, several events of basal immunity were probed. We found that glycyrrhizin induced a transient extracellular alkalinisation, indicative of calcium influx. Furthermore, transcripts of phytoalexins genes, were activated in cells of the grapevine Vitis rupestris, and this induction was followed by accumulation of the glycosylated stilbene alpha-piceid. We also observed that glycyrrhizin was able to induce actin bundling in leaves of a transgenic grape, especially in guard cells. We discuss these data in frame of a model, where glycyrrhizin, through stimulation of RboH, can cause actin remodelling, followed by defence responses, such as calcium influx, induction of phytoalexins transcripts, and accumulation of stilbene glycosides.

Two new hits of triazolyl-indole containing two different alkylsulfanyl analogues named tert-butyl 2-((4-amino-5-(1H-indol-2-yl)-4H-1,2,4-triazol-3-yl)thio)acetate 2 , and ethyl 2-((4-amino-5-(1H-indol-2-yl)-4H-1,2,4-triazol-3-yl)thio)acetate 3 were synthesized via reaction of 4-amino-5-(1H-indol-2-yl)-1,2,4-triazol-3(2H)-thione 1 with tert-butyl bromoacetate and ethyl chloroacetate in the presence of base (Et3N). The molecular structure of 2 , and 3 was confirmed by single-crystal X-ray diffraction and H-1/(CNMR)-C-13 spectroscopic techniques. In compound 2 , the molecular packing depends on significant O center dot center dot center dot H (9.3%), N center dot center dot center dot H (12.4%) and S center dot center dot center dot H (3.1%) as well as relatively weak C center dot center dot center dot H (14.1%), S center dot center dot center dot C (2.1%), H center dot center dot center dot H (50.5%) and S center dot center dot center dot S (0.9%) contacts. Oleanolic acid indole derivatives as novel alpha-glucosidase inhibitors: Synthesis, biological evaluation, and mechanistic analysis

Research efforts have been directed to the development of oleanolic acid (OA) based alpha-glucosidase inhibitors and various OA derivatives showed improved anti-alpha-glucosidase activity. However, the inhibitory effects of indole infused OA derivatives on alpha-glucosidase is unknown. Herein, we synthesized a series of indole-OA (2a-2o) and -OA methyl ester (3a-3 1) derivatives with various electron withdrawing groups inducted to indole benzene ring and evaluated their anti-alpha-glucosidase activity. Indole OA derivatives (2a-2o) exhibited superior alpha-glucosidase inhibitory effects as compared to OA methyl ester derivatives (3a-31) and OA (with IC50 values of 4.02 mu M-5.30 mu M v.s. over 10 mu M and 5.52 mu M, respectively). In addition, mechanistic studies using biochemical (kinetic assay), biophysical (circular dichroism), and computational (docking) methods revealed that OA-indole derivatives (2a and 2f) are mixed type of alpha-glucosidase inhibitors and their inhibitory effects were attributed to their capacity of forming the ligand-enzyme complex with alpha-glucosidase enzyme. Findings from this study support that OA indole derivatives are promising alpha-glucosidase inhibitors as a potential management of diabetes mellitus.
