Reaction of indoles with aromatic fluoromethyl ketones: an efficient synthesis of trifluoromethyl(indolyl)phenylmethanols using K2CO3/n-Bu4PBr in water

A new, mild and efficient protocol for the synthesis of trifluoromethyl(indolyl)phenylmethanols by the reaction of indoles with a variety of aromatic fluoromethyl ketones in the presence of K2CO3 (15 mol %) and n-Bu4PBr (15 mol %) in water. The desired products were obtained in good to excellent yields without requiring a column chromatographic purification. The reusability of the catalytic system and large-scale synthesis of indolyl(phenyl)methanols, which would further transform into biological active indole-derived compounds, are further advantages of this protocol.

Isolation and Identification of Extremophilic Bacteria with Potential as Plant Growth Promoters (Pgpb) of A Geothermal Site: A Case Study

‘Los Negritos is an extremophile geothermal site from Michoacan State, Mexico. Evidence of the diversity of its microbiota and its potential is still lacking. This study aimed to evaluate the bacterial communities by 16S metagenomics of the site and the potential of culturable bacteria isolated from the site by promoting growth plants (PGP) traits. Gemmatimonadetes phylum was the most abundant, followed by Actinobacteria, Alphaproteobacteria, and Firmicutes. A total of 15 culturable strains were isolated and identified. They belong to five different species: Bacillus seohaeanensis, Jeotgalibacillus alkaliphilus, Halobacillus andaensis, Thalassobacillus devorans, and Halobacillus campisalis. The phytohormone indole-3-acetic acid (IAA) production was in the interval from 14.78 to 46.52 mg L-1. The best IAA producers were T. devorans E18 and H. campisalis E20 with tryptophan and H. andaensis, T. devorans, and H. campisalis without tryptophan ranging 39.8 to 51.5 mg L-1. Phosphorous solubilization capability was detected in J. alkaliphilus and T. devorans, whereas B. seohaeanensis was the highest ammonium producer (34.1 +/- 0.52 mg L-1) and H. andaensis was positive for siderophore production. These results demonstrate that, despite the low diversity, bacteria isolated from ‘Los Negritos’ have the potential for plant growth promotion.

Development of Mcl-1 inhibitors for cancer therapy

The myeloid leukemia cell differentiation protein (Mcl-1) is an anti-apoptotic protein of the B-cell lymphoma 2 (Bcl-2) family, which regulates cellular apoptosis. Mcl-1 expression plays a key role in survival of cancer cells and therefore serves as a promising target in cancer therapy. Besides, its importance as a cancer target, various peptides and small-molecule inhibitors have been successfully designed and synthesized, yet no Mcl-1 inhibitor is approved for clinical use. However, recent development on the understanding of Mcl-1’s role in key cellular processes in cancer and an upsurge of reports highlighting its association in various anticancer drug resistance supports the view that Mcl-1 is a key target in various cancers, especially hematological cancers. This review compiles structures of a variety of inhibitors of Mcl-1 reported to date. These include inhibitors based on a diverse range of heterocycles (e.g. indole, imidazole, thiophene, nicotinic acid, piperazine, triazine, thiazole, isoindoline), oligomers (terphenyl, quaterpyridine), polyphenol, phenalene, anthranilic acid, anthraquinone, macrocycles, natural products, and metal-based complexes. In addition, an effort has been made to summarize the structure activity relationships, based on a variety of assays, of some important classes of Mcl-1 inhibitors, giving affinities and selectivities for Mcl-1 compared to other Bcl-2 family members. A focus has been placed on categorizing the inhibitors based on their core frameworks (scaffolds) to appeal to the chemical biologist or medicinal chemist. (C) 2020 The Authors.
