The Molecular docking studies showed that the ligand 1,2,3,4,5 and 6 with better activities have higher bind affinities ranging from (?6.4 and ?10.4 kcal/mol) which formed H-bonds and hydrophobic interactions with amino acid residues of mycobacterum tuberculosis (M. tuberculosis) DNA gyrase receptor. This research has shown that the binding affinities of these inhibitors were found to be better than the commercially sold anti-mycobacterium tuberculosis; enthambutol (?5.8 kcal/mol) and isoniazid (?5.3 kcal/mol). QSAR model generated and molecular docking results propose the direction for the design of new anti-tubercular agents with better activities against DNA gyrase.

In this work, we extended the generic approach for the site-directed immobilization of enzymes based on maleimidethiol coupling of engineered enzymes to the oriented immobilization of variants of bilirubin oxidase from Magnaporthe oryzae (MoBOD) to electrodes. We show that this approach leads to the stable attachment of the enzyme to the electrode surface and that the immobilized MoBOD variants are active for bioelectrocatalytic reduction of dioxygen through direct (unmediated) electron transfer (DET) from the electrode. For the three MoBOD variants studied, significant differences are observed in the kinetics of DET that relate to the orientation of the enzyme and the distance of the T1 site from the electrode surface. The stability of the immobilized enzymes allows us to compare the DET and mediated electron-transfer (MET) pathways and to investigate the effects of pH and Cl-. Our studies show a change in the slope of pH dependence at pH 6.0 and highlight the effect of Cl- on the direct oxygen reduction by MoBOD as a function of pH for the immobilized enzyme and the interconversion of the resting oxidized (RO) form of the immobilized enzyme and the alternative resting (AR) state formed in the presence of Cl-.

Sirtuins (SIRTs) are NAD-dependent deacylases, known to be involved in a variety of pathophysiological processes and thus remain promising therapeutic targets for further validation. Herein, we report an extensive SAR study of this chemical series and identify the key pharmacophoric elements and physiochemical properties that underpin the excellent activity observed. New analogues have been identified with submicromolar SIRT2 inhibtory activity and good to excellent SIRT2 subtype-selectivity. Importantly, we report a cocrystal structure of one of our compounds (29c) bound to SIRT2. This reveals our series to induce the formation of a previously reported selectivity pocket but to bind in an inverted fashion to what might be intuitively expected. We believe these findings will contribute significantly to an understanding of the mechanism of action of SIRT2 inhibitors and to the identification of refined, second generation inhibitors.

Breast cancer is the most prevalent cancer in women. The development of resistances to therapeutic agents and the absence of targeted therapy for triple negative breast cancer motivate the search for alternative treatments. With this aim in mind, we synthesised new derivatives of troglitazone, a compound which was formerly used as an anti-diabetic agent and which exhibits anti-proliferative activity on various cancer cell lines. Among the compounds prepared, some displayed micromolar activity against hormone-dependent and hormone-independent breast cancer cells. Furthermore, the influence of the compounds on the viability of primary cultures of human hepatocytes was evaluated. This enabled us to obtain for the first time interesting structure-toxicity relationships in this family of compounds, resulting in 6b and 8b, which show good anti-proliferative activities and poor toxicity towards hepatocytes, compared to troglitazone.
