In the title pyrazoline derivative, C16H16N2O3 center dot H2O, the pyrazoline ring has an envelope conformation with the substituted sp(2) C atom on the flap. The pyrazoline ring makes angles of 86.73 (12) and 13.44 (12)degrees with the trisubstituted and disubstituted benzene rings, respectively. In the crystal structure, the molecules are connected into chains running in the b-axis direction by O-H center dot center dot center dot N hydrogen bonding. Parallel chains interact through N-H center dot center dot center dot O hydrogen bonds and pi-pi stacking of the trisubstituted phenyl rings. One new diarylheptanoid (1) and one previously unknown ascorbic acid derivative (2) along with 48 known compounds including three ascorbic acid derivatives (3-5), three diarylheptanoids (6-8), three juglone derivates (9-11), six racemic alpha-tetralones (12a/b-17a/b), four anthraquinones (18-21), eight flavonoids (22-29), nine oligomeric proanthocyanidins (30-38), one phenolic aldehyde (39), one aromatic ketone (40), one pyrrole alkaloid (41), one steroid (42), six terpenes (43-48) and two fatty acids (49-50) were isolated from the leaves of Juglans regia. Historical stringed musical instruments are a unique class of cultural heritage objects. Crafted during the 16th-18th centuries, these instruments remain somewhat mysterious. Due to the absence of written historical documents of traditional varnish recipes, their chemical characterization is the only way to recover the lost ‘secret’ of the Cremonese stringed instrument maker Antonio Stradivari and his contemporaries, whose traditions had been lost by 1800. Pyrolysis combined with gas chromatography-mass spectrometry (GC-MS) using solid-phase microextraction (SPME) followed by on-fiber silylation (Py SPME Syl GC-MS) was applied to a large collection of standards to evaluate its validity for compositional analysis and material identification of historical stringed instrument varnishes. The standards included pure resins (colophony, amber, mastic, sandarac, elemi), different types of shellac, as well as colorants or additives such as madder, dragon’s blood, and Aloe vera powder, and other materials (propolis, beeswax). The aim was to obtain an extensive collection of GC-MS data of silylated pyrolysis products useful for the characterization of the finishes of these unique historical objects. A set of distinct pyrolysis patterns and products were chosen as specific molecular markers for each standard. The method was applied to instrument varnish samples from Stainer, Baioni, Amati, Guarneri, Gasparo da Salo, Maggini, and Guadagnini instruments. The method was shown to be effective for the identification of oxidized lignin, drying oils, and diterpenic resins. In the case of dragon’s blood, madder, shellac, and propolis new markers were identified, suggesting their presence in some instrument varnishes. The identification of proteins and madder appeared most challenging.

The current chemotherapy against Chagas disease is inadequate and insufficient. A series of ten Mannich base-type derivatives have been synthesized to evaluate their in vitro antichagasic activity. After a preliminary screening, compounds 7 and 9 were subjected to in vivo assays in a murine model. Both compounds caused a substantial decrease in parasitemia in the chronic phase, which was an even better result than that of the reference drug benznidazole. In addition, compound 9 also showed better antichagasic activity during the acute phase. Moreover, metabolite excretion, effect on mitochondrial membrane potential and the inhibition of superoxide dismutase (SOD) studies were also performed to identify their possible mechanism of action. Finally, docking studies proposed a binding mode of the Fe-SOD enzyme similar to our previous series, which validated our design strategy. Therefore, the results suggest that these compounds should be considered for further preclinical evaluation as antichagasic agents.

Monoclonal antibodies (mAb) are a major focus of the pharmaceutical industry, and polyclonal immunoglobulin G (IgG) therapy is used to treat a wide variety of health conditions. As some individuals require mAb/IgG therapy their entire life, there is currently a great desire to formulate antibodies for bolus injection rather than infusion. However, to achieve the required doses, very concentrated antibody solutions may be required. Unfortunately, mAb/IgG self-assembly at high concentration can produce an unacceptably high viscosity for injection. To address this challenge, this study expands the concept of dynamic covalent chemistry to dynamic bioconjugation in order to reduce viscosity by interfering with antibody-antibody interactions. Ruthenium complexes(1-3) have been synthesized using pincer-type ligands L-1 = (E)-2-((2-phenyl-2-(pyridin-2-yl)hydrazono)methyl)pyridine, L-2 = (E)-2-(1-phenyl-2-(1-(pyridin-2-yl)ethylidene)hydrazinyl)pyridine, L-3 = (E)-2-(phenyl(2-phenyl-2-(pyridin-2-yl)hydrazono) methyl)pyridine. The molecular structures of all the complexes 1, 2 and 3 were determined by using single crystal X-ray diffraction. These complexes showed excellent catalytic activities such as transfer hydrogenation and alcohol oxidation. Lung cancer is a leading cause of cancer-related death worldwide. Cyanopyridines and aminocyanopyridines with carbon-nitrogen bonds have been proved to exert significant anticancer, antibacterial, and anti-inflammatory effects. In this study, we showed that aminocyanopyridine 3o and 3k displaying potent antitumor activity via inhibiting the signal transducer and activator of transcription 3 (STAT3) pathway. They blocked the constitutive STAT3 phosphorylation in a dose- and time-dependent manner and regulated the transcription of STAT3 target genes encoding apoptosis factors. Most importantly, 3o also inhibited interleukin-6-induced STAT3 activation and nuclear localization. Furthermore, 3o significantly inhibited the tumor growth of H460-derived xenografts. Taken together, these findings suggest that 3o and 3k are promising therapeutic drug candidates for lung cancer by inhibiting persistent STAT3 signaling.
