Using thioglycosides as donors and trimethylsilyl trifluoromethanesulfonate (TMSOTf) and N-iodosuccinimide (NIS) as promoter, a modified procedure for the glycosylation of spirostanols was developed. A series of saponins were synthesized in mild condition with excellent yields. E.g., sapogenin glucoside I (R = phthalimido) was prepared with quant. yield by reacting diosgenin with Et 3,4,6-tri-O-acetyl-2-deoxy-2-phthalimido-1-thio-β-D-glucopyranoside using TMSOTf, NIS and 4Å mol. sieves in CH2Cl2 at -15°.
